坏损外担菌(Exobasidium vexans)植物生长物质种类及含量研究 被引量:5

摘要 为研究坏损外担菌(Exobasidium vexans)培养液中植物生长物质种类及含量。建立HPLC法测定坏损外担菌(E.vexans)培养液中玉米素(Z)、赤霉素(GA3)、吲哚乙酸(IAA)、3-吲哚丁酸(IBA)、激动素(KT)、脱落酸(ABA)、和6-苄氨基嘌呤(6-BA)含量。流动相A:甲醇∶乙腈∶水=5∶1∶17;流动相B:甲醇∶乙腈∶水=6∶3∶8,色谱条件为:Waters C18反相色谱柱(4.6 mm×250mm,5μm),柱温35℃、流速0.8 m L·min-1,检测波长为254 nm,进样体积10μL,分别测定培养1、3、5、7、9、11、13 d时培养液生长物质含量。结果表明:坏损外担菌(E.vexans)培养液中检测出5种植物生长物质。培养到5 d时,玉米素和赤霉素含量达到最高值;培养到7 d时,激动素、6-苄氨基嘌呤和3-吲哚丁酸含量达到最高值。 In order to study the species and content of plant growth material in culture medium of Exobasidium vexans, a high performance liquid chromatography ( HPLC) method was established for the determination of ZA, gibberellin (GA3), indole acetic acid (IAA), 3 -indole butyric acid (IBA), kinetin (KT), abscisic acid (ABA), and 6 -benzylaminopurine (6 -BA). Mobile phase A: methanol ∶acetonitrile ∶water = 5 ∶1 ∶17, mobile phase B: methanol ∶acetonitrile ∶water = 6 ∶3 ∶8, the chromatographic conditions were as followings: Waters C18 column (4.6 mm×250 mm,5 μm), column temperature 35 ℃, flow rate 0.8 mL min -1 , detection wavelength 254 nm, sample volume 10 μL. The contents of growth medium in culture medium were measured at 1, 3, 5, 7, 9, 11 and 13 d. Under the experimental conditions, five kinds of plant growth substances were detected in E. vexans liquid culture medium. The content of zeatin and gibberellin reached the highest level on the 5th day. The content of zeatin, 6 -benzylaminopurine and 3 -indole butyric acid reached the highest level on the 7th day.
