个人用葡萄糖传感器(PGM)即时检测金黄色葡萄球菌及血浆凝固酶鉴定 被引量:1

PGM-based point-of-care testing of S.aureus and coagulase identification

摘要 建立基于个人用葡萄糖传感器(personal glucose meter,PGM)的血浆凝固酶阳性金黄色葡萄球菌(Staphylococus aureus,S.aureus)即时检测(point-of-care testing,POCT)方法,将定量和血浆凝固酶试验合并为一步完成,达到快速、简便、经济和准确的目的,便于推广应用。通过优化配方得到一种专用于PGM检测的Baird Parker-BHI-Glu混合培养基,并建立一套标准的检测方法;同时从自然界中分离纯化多株野生S.aureus;用加标回收法验证该方法与国标法在定量和血浆凝固酶试验上的符合程度,确定其可行性。结果显示:本试验成功设计优化了PGM即时检测血浆凝固酶阳性S.aureus的专用培养基,建立了检测标准方法,并制作了标准曲线,检测限为5.9×10^0CFU/mL,加标样品检出率及血浆凝固酶阳性样品检出率分别为100.00%和96.55%,检测时间不超过24h,为实际应用与商品化提供了理论支撑。 A innovative method in this study used personal glucose meter(PGM) based point-of-care testing(POCT) to test coagulase-positive S. aureus,which integrates the quantitative test with co- agulase test into one step and has been proved to be an accurate, time-efficient and cost-effective advantage. Basically,optimizing the original recipe obtained a Baird Parker-BHI Glu medium mixture dedicated for PGM POCT method only and developing standard testing procedures corre- spondingly. In order to further verification the wild S. aureus was separated and purified from nature environment, and sample recovery method was used to examine the conformity of results (from quantitative and coagulase tests) between PGM POCT and the national standard method. According to the experiment results,a standard curve was also attained with the detection limit be- ing 5.9 × 10^0 CFU/mL and the detection rate of positive sample and coagulase-positive sample be- ing 100.00% and 96.55% respectively. It is noteworthy to mention that the detection time for our experiment was within 24 hours. The establishment of PGM-based POCT method for coagulasepositive S. aureus can provide a technique foundation for practical application and commercialization.
