摘要
通过使用双光子显微镜活体成像技术优化了K/B×N血清诱导的关节炎小鼠模型,并观察了嗜中性粒细胞的募集情况。皮下注射K/B×N血清可以快速诱导大量的嗜中性粒细胞募集到局部炎症关节。通过使用这种方法,发现阻断C5a活性会减少嗜中性粒细胞募集,说明C5a是关节炎期间嗜中性粒细胞募集的关键调节因子。因此,本试验将为补体相关生物制剂治疗类风湿关节炎提供新的靶点。
Rheumatoid arthritis (RA) is a common disease that causes deformities,but its patholo- gy is not fully clear. The infiltration of neutrophils is a prominent feature of inflammatory diseases including rheumatoid arthritis. Understanding how neutrophil recruitment is regulated during pathogenesis is crucial for developing anti-inflammatory drugs. In this study, we optimized the K/B × N serum-induced mouse arthritis model to observe the recruitment of neutrophil in vivq using two-photon microscopy. K/B × N arthritogenic serum was injected subcutaneously into ode hhad footpad to rapidly induce a local arthritis with robust neutrophil recruitment. Using this approach, we showed that the blockage of CSa activity with monoclonal antibody impaired neutrophil recruitment and demonstrated the critic role of C5a in initiation. Therefore, these findings will provide regulating neutrophil recruitment during arthritis a new target for the treatment of rheumatoid arthritis with complement-related biological agents.
