阿霉素化疗和槲皮素对大鼠肾脏中间丝蛋白表达及肾脏炎症反应的影响 被引量:1

Effects of Adriamycin Chemotherapy and Quercetin on Intermediate Filament Protein Expression and Kidney Inflammation in Rats

摘要 为了揭示阿霉素化疗和槲皮素给药对大鼠肾脏中间丝蛋白的表达及肾脏炎症反应的影响,并探讨槲皮素对阿霉素诱导的肾毒性的预防作用。将50只雄性SD大鼠分为5组(每组10只):对照组、阿霉素处理组(剂量为20 mg/kg bw)、槲皮素治疗组(剂量为50 mg/kg bw)、槲皮素+阿霉素组和阿霉素+槲皮素组,测量各组大鼠生化参数和肾功能。此外,对肾组织炎症标记物进行评估,并采用实时定量PCR分析中间丝蛋白mRNA表达情况。结果显示,阿霉素治疗前和治疗中应用槲皮素治疗,可有效减缓由阿霉素诱导的肾毒性,并显著改善肾功能。槲皮素通过调节中间丝蛋白mRNA的表达来抑制炎症反应并保护足细胞免受损伤。因此,槲皮素除了具有治疗作用之外,还可以作为预防剂在临床化疗中抑制阿霉素诱导的肾毒性。 In order to reveal the effect of adriamycin chemotherapy and quercetin administration on the expression of intermediate filament protein and renal inflammatory response in rats, and to explore the preventive effect of quercetin on adriamycin-induced nephrotoxicity. 50 male SD rats were divided into 5 groups (10 per group): control group, doxorubicin-treated group (dose of 20 mg/kg bw), quercetin-treated group (dose of 50 mg/kg bw), quercetin+ adriamycin group and doxorubicin+quercetin group, biochemical parameters and renal function in each group were measured. In addition, renal inflammation markers were evaluated and real-time quantitative PCR was used to analyze mRNA expression of intermediate filament protein. The results showed that treatment with quercetin before and during treatment with doxorubicin can effectively reduce the nephrotoxicity induced by doxorubicin, and significantly improve renal function. Quercetin inhibits inflammatory responses and protects podocytes from damage by regulating the expression of intermediate filament protein mRNA. Therefore, in addition to its therapeutic effect, quercetin can also be used as a prophylactic agent to inhibit doxorubicin-induced nephrotoxicity in clinical chemotherapy.
