疏肝泻火方对雌性性早熟大鼠血清IGF-1、GH、BGP水平的影响

Effects of Shugan Xiehuo Formula on Serum Levels of IGF-1,GH and BGP in Female Rats with Precocious Puberty

目的：观察疏肝泻火方对雌性性早熟大鼠身长、胫骨长度和血清胰岛素样生长因子-1（IGF-1）、生长激素（GH）、骨钙素（BGP）水平的影响。方法：取SD雌性大鼠60只,随机分为正常对照组、性早熟空白组、亮丙瑞林组以及疏肝泻火高、中、低剂量组6组,除正常对照组外,其余组大鼠采用兴奋性氨基酸-N-甲基-DL-天冬氨酸（NMA）诱导建立性早熟模型,并分别予药物干预,观察大鼠顶臀径、胫骨长度及黄体出现情况,测定血清IGF-1、GH、BGP水平。结果：与性早熟空白组比较,正常对照组、亮丙瑞林组、疏肝泻火高剂量组大鼠顶臀径较小（P〈0.05）;与性早熟空白组比较,亮丙瑞林组与疏肝泻火高、中剂量组血清GH、IGF-1、BGF的浓度均较低（P〈0.05）,低剂量组血清GH、IGF-1浓度较低（P〈0.05）,但高于亮丙瑞林组（P〈0.05）。结论：疏肝泻火方改善性早熟患儿成年身高的机制可能与降低血清GH、IGF-1、BGF的浓度有关,且治疗效果与疏肝泻火方剂量相关。

Objective：To observe the effect of Shugan Xiehuo Formula on the body length,tibia length and the levels of serum insulin like growth factor-1（IGF-1）,growth hormone（GH） and osteocalcin（BGP） in the female precocious rats. Methods：Total 60 female SD rats were randomly divided into 6 groups：the normal control group,precocious blank group,leuprorelin group and high,medium and low dose group of Shugan Xiehuo Formula. Except the normal control group,the precocious model was induced by NMA,and the top hip diameter,the length of tibia and the appearance of corpus luteum in the rats were observed and the serum levels of IGF-1,GH and BGP were measured. Results：Compared with the precocious blank group,the top hip diameter of the normal control group,leuproprolin group and the high dose group of Shugan Xiehuo Formula were smaller（P〈0.05）. The concentration of GH,IGF-1 and BGF in the serum of the leuprorelin group,the high and the medium dose groups of Shugan Xiehuo Formula were lower than that in precocious blank group（P〈0.05）,and the serum GH and IGF-1 concentrations in the low dose group was lower too（P〈0.05）,but it was higher than that in the leuprorelin group（P〈0.05）. Conclusion：The mechanism of improving adult height in children with precocious puberty by Shugan Xiehuo Formula may be related to reduce the concentration of serum levels of GH,IGF-1 and BGF,and the therapeutic effect is related to the dosage of Shugan Xiehuo Formula.
