水飞蓟宾对前列腺癌细胞增殖、凋亡的影响及其作用机制 被引量:7

摘要 目的研究水飞蓟宾对前列腺癌细胞增殖、凋亡的影响,并探讨其分子生物学机制。方法使用前列腺癌细胞系C4-2和PC-3作为研究对象。采用不同浓度的水飞蓟宾处理前列腺癌细胞,应用MTT实验检测水飞蓟宾对于前列腺癌细胞增殖活性的影响;应用平板克隆实验检测水飞蓟宾对前列腺癌细胞增殖能力影响;应用蛋白免疫印迹(Western blot)方法测定水飞蓟宾处理后前列腺癌细胞Caspase 3/Cleaved-Caspase 3、Caspase 12/Cleaved-Caspase 12、BiP/GRP78、IRE1、JNK蛋白水平改变。结果MTT实验发现水飞蓟宾抑制前列腺癌细胞增殖,呈浓度依赖性;平板克隆实验显示水飞蓟宾抑制前列腺癌细胞增殖能力;Western blot发现水飞蓟宾处理后,凋亡相关蛋白Cleaved-Caspase3和Cleaved-Caspase12表达升高,内质网应激相关蛋白BiP、IRE1和JNK上调,并呈浓度依赖性。结论水飞蓟宾通过激活内质网应激抑制前列腺癌细胞增殖、促进凋亡。 Objective To explore the effects and mechanism of silibinin on the proliferation and apoptosis of human prostate cancer cells. Methods Human prostate cancer C4-2 and PC-3 cells were treated with different concentrations of silibinin.The cellular viabilities of C4-2 and PC-3 cells were determined with MTT.The proliferation abilities of C4-2 and PC-3 cells were detected with cell colony tablet cloning assay.The protein levels of Caspase 3/Cleaved-Caspase 3,Caspase 12/Cleaved-Caspase 12,BiP,IRE1 and JNK were tested with Western blotting. Results Silibinin inhibited the proliferation of C4-2 and PC-3 cells in a concentration-dependent manner.After silibinin treatment,the expressions of Cleaved-Caspase 3,Cleaved-Caspase 12,BiP,IRE1 and JNK upregulated. Conclusion Silibinin can inhibit the proliferation and apoptosis of human prostate cancer cells by activating endoplasmic reticulum stress.
