双丹明目胶囊对糖尿病大鼠模型视网膜组织结构改变的影响 被引量:3

摘要 目的观察双丹明目胶囊对糖尿病大鼠模型视网膜组织结构的影响。方法将40只雄性SD大鼠,采用随机数字法分为空白组(A组)、模型组(B组)、双丹明目组(C组)、阳性对照组(D组)4组,每组10只20眼。将B、C、D组实验大鼠采用链脲佐菌素(STZ)50mg/kg的剂量一次性大鼠尾静脉注射法建立糖尿病视网膜病变大鼠模型,造模后1周开始连续灌胃用药,灌胃后4周、8周,每组分别处死一半动物(各5只大鼠),光镜和透射电镜观察视网膜组织形态。结果造模后用药第4周、8周,双丹明目组、阳性对照组的大鼠视网膜组织形态和超微组织结构改变明显好于模型组,其结构层次清晰,仅组织轻度水肿,部分毛细血管内皮细胞轻度增生;而模型组大鼠视网膜组织水肿明显,毛细血管壁增厚,内皮细胞增生,纤维组织增生明显;超微组织结构显示,模型组大鼠视网膜毛细血管内皮细胞明显肿胀,胞体变圆,突向管腔,线粒体肿胀,空泡化,基底膜增厚较明显。结论糖尿病模型大鼠视网膜组织水肿明显,毛细血管壁增厚,内皮细胞增生,纤维组织增生明显;双丹明目胶囊能明显改善糖尿病大鼠模型视网膜组织形态和超微组织结构。 OBJECTIVE To study the effects of Shuangdan Mingmu capsule on the changes of retina tissue structure in a diabetic rat model. METHODS Forty male SD rats were divided into blank group(A), model group(B), Shuangdan Mingmu capsule group(C) and positive control group(D) using random number method(10 rats with 20 eyes per group). A rat model of diabetic retinopathy(DR) was established with the disposable injection of Streptozotocin(STZ) at dose of 50 mg/kg through rat tail vein in experimental rats in group B, group C and group D. Intragastric administration was performed 1 week after the model was established, 5 rats per group were sacrificed 4 weeks and 8 weeks after intragastric administration, respectively. The morphology of retina was observed by light microscope and electron microscope. RESULTS At the 4 th and 8 th week of intragastric administration after modeling, compared with the model group, the morphological and the ultrastructure changes of the retinal tissues were improved obviously in the Shuangdan Mingmu capsule group and the positive control group. Specifically, they were of clear structure layer, mild tissue edema and partial mild-ly-proliferated capillary endothelial cells. Meanwhile, obvious edema, thickened capillary wall,proliferated endothelial cells and obvious proliferation of fibrous tissue were found in the retina tissues of rats in the model group; and the ultrastructure results showed that the retinal capillary endothelial cells of rats in the model group were obviously swollen, with cell body became round and protruding toward the lumen, swollen and vacuolation mitochondria, and basement membrane were obvious thickened. CONCLUSIONS Obvious edema, capillary wall thickening, endothelial cell proliferation and obvious proliferation of fibrous tissue were observed in retina tissues of rats in the DR model; and Shuangdan Mingmu capsule significantly improved the morphology and ultra-structure of the retinal tissues in DR rat model.
