MiRNA-375表达对非小细胞肺癌侵袭和转移影响

EFFECT OF MIRNA-375 ON INVASION AND METASTASIS OF NON-SMALL CELL LUNG CANCER

目的探讨miRNA-375(miR-375)对非小细胞肺癌(NSCLC)侵袭迁移的影响并探讨其机制。方法用空载体慢病毒转染NSCLC细胞(A549)作为对照组,用慢病毒miR-375载体转染A549细胞作为实验组。利用荧光定量PCR技术检测miR-375的表达,利用Transwell实验检测侵袭和转移能力的变化,利用Western blot技术检测CIP2A蛋白表达量的变化。结果miR-375在NSCLC细胞中呈高表达。与对照组相比,实验组在上调miR-375后,细胞的侵袭和转移能力增加,差异均有显著性(t=4.41、4.57,P<0.05);同时CIP2A蛋白的表达量也明显升高,差异有显著性(t=4.57,P<0.05)。结论 miR-375高表达可增强NSCLC细胞的侵袭和转移能力,其机制可能通过上调CIP2A蛋白的表达而实现。

Objective To investigate the effect of miRNA-375 （miR-375） on the invasion and metastasis of non-small cell lung cancer （NSCLC） and the related mechanism. Methods NSCLC A549 cells transfected with empty vector lentivirus were established as control group, and those transfected with lentiviral miR-375 vector were established as experimental group. Quantitative real-time PCR was used to measure the expression of miR-375, Transwell assay was used to evaluate the changes in invasion and metastasis abilities, and Western blot was used to measure the change in the protein expression of CIP2A. Results The results showed that miR-375 was highly expressed in NSCLC. Compared with the control group, the experimental group had significant increases in invasion and metastasis abilities （ t =4.41 and 4.57, P 〈0.05） and the protein expression of CIP2A （ t =4.57, P 〈0.05） after the upregulation of miR-375. Conclusion High expression of miR-375 can promote the invasion and metastasis abilities of NSCLC cells, possibly by upregulating the protein expression of CIP2A.
