HOXD3在胃癌组织中的表达及其临床意义 被引量:1

EXPRESSION OF HOXD3 IN GASTRIC CANCER TISSUE AND ITS CLINICAL SIGNIFICANCE

摘要 目的探究HOXD3在胃癌组织中的表达情况及其临床意义。方法收集我院普外科2010年6月—2012年9月收治的76例胃癌病人肿瘤组织及相应癌旁组织标本,并收集病人相应的临床病理资料及随访资料,采用荧光定量链式聚合酶链反应技术及免疫组织化学染色技术检测肿瘤组织及癌旁组织HOXD3mRNA表达及蛋白阳性率,分析HOXD3表达与病人临床病理参数及预后间的关系。结果 HOXD3mRNA在胃癌组织中表达水平显著高于癌旁组织(t=10.48,P<0.01),HOXD3蛋白在胃癌组织中阳性率也显著高于癌旁组织(χ~2=23.30,P<0.01),且HOXD3蛋白阳性病人的TNM分期较高、肿瘤组织分化程度及Lauren分型不良,HOXD3mRNA表达与病人TNM分期及肿瘤组织分化程度呈显著相关(χ~2=5.217、9.002,r=0.406、0.540,P<0.05)。HOXD3阳性病人总生存率及无病生存率相比阴性病人显著下降(HR=1.368、1.452,P<0.05)。结论 HOXD3在胃癌组织中高表达,且与胃癌的发生发展及病人预后密切相关,是潜在的分子靶向位点。 Objective To investigate the expression of HOXD3 in gastric cancer tissue and its clinical significance.Methods A total of 76 patients with gastric cancer who were admitted to Department of General Surgery in our hospital form June 2010 to September 2012 were enrolled, and gastric cancer tissue samples and corresponding adjacent tissue samples were collected. Their clinicopathological data and follow-up data were also collected. Quantitative real-time PCR and immunohistochemical staining were used to measure the mRNA expression of HOXD3 and the positive rate of HOXD3 protein. The association of HOXD3 expression with clinicopathological parameters and prognosis was analyzed. Results Compared with the adjacent tissue, the gastric cancer tissue had significantly higher mRNA expression of HOXD3 ( t =10.48, P 〈0.01) and positive rate of HOXD3 protein ( χ 2= 23.30, P 〈0.01). The patients with positive HOXD3 protein had a high TNM stage, a high degree of tumor differentiation, and a poor Lauren type, and the mRNA expression of HOXD3 was significantly correlated with TNM stage and degree of tumor differentiation ( χ^2=5.217 and 9.002, r =0.406 and 0.540, P 〈0.05). The patients with positive HOXD3 had significantly lower overall survival rate and disease-free survival rate than those with negative HOXD3 (hazard ratio=1.368 and 1.452, P 〈0.05).Conclusion HOXD3 is highly expressed in gastric cancer tissue and is closely associated with the development, progression, and prognosis of gastric cancer. Therefore, it is a potential molecular target site.
