摘要
为了对青海互助北山林区冬瓜杨遗产多样性进行测定,采集青海互助北山林区不同地理居群冬瓜杨叶片共20个样品,并进行生物学鉴定.在本研究中青海互助北山林区冬瓜杨ITS序列的扩增片片段在700~800bp间,单倍型多态性水平为0.6879,基因流Nm(0.0486)<1.青海互助北山林区不同地理居群冬瓜杨ITS序列的扩增片段满足杨属DNA条形码的片段,psbA-trnH序列分析,高于植物平均水平,但是青海互助北山林区冬瓜杨基因交流程度较低,因而,居群之间的遗传变异较大.
In order to determine the heritage diversity of populous purdomii in the main forest areas of Qinghai,20 samples were collected from the leaves of populous purdomii from different geographical populations in the Beishan Forest region of Huzhu,Qinghai Province,and their biological identification was carried out.In this study,the amplified fragment of ITS sequence of populous purdomii in Huzhu Mountain Forest region of Qinghai Province was between 700~800bp,the level of haplotype polymorphism was 0.6879 ,and the gene flow was(0.0486) 〈 1.The amplified ITS sequence of populous purdomii from different geographical populations in Huzhu Mountain Forest area of Qinghai Province met the requirements of DNA barcode sequence analysis of poplar,which was higher than the plant average level,but the sequence was higher than the average level in Qinghai Province.In Beishan forest area,the degree of gene exchange was low,so the genetic variation among populations was higher.
