半夏生物碱对肺上皮细胞炎症损伤的保护作用研究 被引量:18

RESEARCH ON THE PROTECTIVE EFFECT OF ALKALOIDS Pinelliaf Rhizoma ON INFLAMMATORY INJURY OF LUNG EPITHELIAL CELLS

摘要 目的:探讨半夏生物碱对肺上皮细胞炎症损伤的保护作用。方法:通过脂多糖(LPS)诱导肺上皮细胞(A549细胞)构建肺上皮细胞炎症损伤的体外模型,CCK-8法检测细胞活性;Griess试剂法测定NO释放量,酶联免疫吸附测定(ELISA)法检测上清液中TNF-α、IL-8、ICAM-1水平;Real time-PCR法检测IL-8、ICAM-1 mRNA表达。结果:半夏生物碱能保护LPS诱导的A549细胞炎症损伤(P<0.05),降低炎症模型细胞NO、TNF-α、IL-8、ICAM-1的释放(P<0.05),抑制IL-8、ICAM-1 mRNA的表达(P<0.05);结论:半夏生物碱可有效保护肺上皮细胞的的炎症损伤,其机制可能与抑制NO、TNF-α能炎症因子的释放有关,还可通过抑制IL-8、ICAM-1的表达,减缓其中性粒细胞的趋化作用,有利于急性肺损伤的早期防治。 Objective To investigate the protective effect of alkaloids from Pinelliae Rhizoma on the inflammation damage of lung epithelial cells. Methods The model of inflammatory injury in lung epithelial cells( A549 cells) was induced by Lipopolysaccharide( LPS) in vitro. Cell viability was assayed by CCK-8,the quantity of NO production was measured by Griess reagent,levels of cytokines tumor necrosis factor-α( TNF-α),interleukin 8( IL-8) and intercellular cell adhesion molecule-1( ICAM-1) were detected by enzyme linked immunosorbent assay( ELISA),and the expression of IL-8 and ICAM-1 mRNA were analyzed by real-time PCR. Results The alkaloids from Pinelliae Rhizoma can effectively protect the A549 cells induced by LPS( P 〈0. 05),and can significantly inhibit the production of NO,TNF-α,IL-8 and ICAM-1( P 〈0. 05),inhibit the expression of IL-8 and ICAM-1 mRNA( P 〈0. 05). Conclusion The alkaloids from Pinelliae Rhizoma have certain protective effect on pulmonary epithelial cells induced by LPS,which may be related to its ability to inhibit the release of NO、TNF-α. The alkaloids can also inhibit the expression of IL-8 and ICAM-1,which may be help to alleviate the chemotaxis of neutrophils induced by IL-8 and ICAM-1.
