黄芪散对T2DM大鼠体脂系数和脂肪组织MCP-1、UCP-1、PRDM16、Cidea mRNA表达的影响 被引量:4

Effects of Huangqi Powder on Fat of Rats Fed with Glucocorticoid and High Fat Diet

摘要 目的:采用糖皮质激素联合高脂喂养建立2型糖尿病(T2DM)大鼠模型,探讨黄芪散对T2DM大鼠体脂系数(肾周脂肪、附睾脂肪和棕色脂肪)及相关基因MCP-1、UCP-1、PRDM16、Cidea mRNA表达的影响。方法:SD大鼠适应性饲养1周后,随机分为正常组(Nor)、糖皮质激素+高脂模型组(HFD+GC),黄芪散组(HQS),每组10只。正常组喂基础饲料,其他两组喂高脂饲料。同时除正常组给予等体积的生理盐水外,其他两组大鼠灌胃醋酸泼尼松3. 5 mg/kg,1次/d,给药组1 h后灌胃黄芪散2. 96 g/kg。于给药12周后,测定各组大鼠空腹血糖(FBG),分离肾周、附睾周围组织的白色脂肪,分离背部颈下肩胛间区中间棕色脂肪,称重,计算体脂系数。采用real-time PCR法检测白色脂肪组织中单核细胞趋化蛋白-1(MCP-1)和棕色脂肪组织中解偶联蛋白(UCP-1)、PR域包含16(PRDM16)、细胞死亡诱导DNA断裂因子α类似效应A(Cidea) mRNA表达的影响。结果:与正常组比较,HFD+GC模型组大鼠表现为高血糖、肾周脂肪系数、附睾脂肪系数显著性升高,MCP-1上调明显,棕色脂肪系数下降,UCP-1、PRDM16、Cidea表达降低。与模型组比较,HQS可降低大鼠白色脂肪系数及MCP-1表达,升高棕色脂肪系数及UCP-1、Cidea的表达。结论:黄芪散可减少T2DM模型大鼠伴随的肥胖症状,可提高机体能量代谢,可作用可有与降低MCP-1表达,升高UCP-1、Cidea表达有关。 Objective: To observe the effects of Huangqi San on body fat coefficient( including perirenal fat,epididymal fat and brown fat) and related genes mRNA expressions( including MCP-1,UCP-1,PRDM16,Cidea) of T2 DM rats induced by glucocorticoid and high fat diet. Method: The study was performed after the rats received adaptive feeding for1 week. Rats were randomly divided into normal control group( Nor),high fat diet with glucocorticoid group( HFD +GC),Huangqi San group( HQS),10 rats in each group. The rats in Nor group was fed with basic diet,and other groups were fed with high-fat diet. At the same time,the rats in the other two groups were given prednisone acetate 3. 5 mg/kg,1 times/d. The normal group was given the normal saline of equal volume. After 1 h in the administration group,Huangqi San 2. 96 g/kg was taken. In 12 weeks after administration,the rats were measured fasting blood glucose(FBG) to seperate white fat tissue from epididymal tissue and perrirenal fat and seperate brown fat tissue from shoulder blades of back. The body fat rate was calculated. Monocyte chemoattractant protein-1( MCP-1) in white adipose tissue was detected by real-time PCR method and uncoupling protein( UCP-1) in the brown adipose tissue,PR domain containing 16( PRDM16),cell death induced by DNA fragmentation factor alpha A( Cidea) in brown adipose tissue were also detected by real-time PCR method. Results: As compared with the normal group,HFD + GC model group ratsshowed hyperglycemia,and increased perirenal fat coefficient,epididymal fat coefficient and expression of MCP-1,decreased brown fat coefficient and mRNA expressions of UCP-1 and Cidea. Conclusion: HQS could reduce the obesity symptoms associated with T2 DM model rats and can increase energy metabolism in rats,which may be related to lowering the expression of MCP-1 and increasing the expressions of UCP-1 and Cidea.
