基于BDNF蛋白表达研究电针俞募穴对便秘大鼠脑肠交互作用的影响机制 被引量:8

摘要 目的:基于BDNF蛋白表达探讨电针大肠俞募穴治疗功能性便秘的脑肠交互作用机制。方法:35只大鼠随机分为空白组、模型组、俞穴组(大肠俞)、募穴组(天枢)和俞募组(大肠俞+天枢)。复方地芬诺酯灌胃造大鼠便秘模型;针刺组每次电针20 min,每天1次,连续治疗10次。运用免疫组化观察针刺治疗前后大鼠海马和结肠组织BDNF蛋白表达情况。结果:各组大鼠海马的CA1、CA2、CA3和DG区BDNF蛋白含量无明显差异(P〉0. 05)。模型组结肠组织BDNF蛋白与空白组比较无显著性差异(P〉 0. 05),针刺组与模型组相比,BDNF含量均升高,以募穴组差异最明显(P 〈0. 01)。结论:电针大肠俞募穴促进排便可能是通过提高结肠组织BDNF含量来实现。 Objective: To explore the mechanism of brain intestinal interaction in treating functional constipation by electroacupuncture at Shu-Mu pointsbased on BDNF expression. Methods: Thirty-five rats were randomly divided into blank group,model group,back-Shu group( Dachangshu),front-Mu group( Tianshu) and Shu-Mu group(Dachangshu + Tianshu). Compound diphenoxylate tablets were gavaged for modeing. The acupuncture group was treated with electroacupuncture at 20 min and once a day for 10 times. The expression of BDNF in hippocampal and colon tissues of rats was observed by immunohistochemical. Results: The contents of BDNF in CA1,CA2,CA3 and DG of hippocampus were not significantly different( P〉0. 05). There was no significant difference in BDNF between the model group and the blank group( P〉0. 05). Compared with the model group,the content of BDNF was increased,and the difference was most obvious in the front-Mu group( P〈0. 01). Conclusion: Electroacupuncture at Shu-Mu points to promote defecation may be achieved by increasing the content of BDNF in colon tissue.
