摘要
目的通过采用气相色谱-质谱法(GC-MS)结合代谢组学方法以阐明南方"傣族"少数民族治疗慢性支气管炎和炎症的民间药物黑面神(大戟科)对大鼠肝肾机体代谢的影响。方法将大鼠分为对照组、黑面神(2 g/kg)组和10%酒精处理组,连续给药7 d。第8 d,从3组大鼠收集肝脏和肾脏样本,衍生化处理后,进行GC-MS分析。结果根据肝脏和肾脏的代谢组学偏最小二乘法鉴别分析(PLS-DA),结果显示黑面神组与对照组和10%酒精处理组区别开来。在肝组织中,黑面神组与对照组之间没有发现差异,而黑面神组与10%乙醇处理组之间的核糖醇和D-葡萄糖含量差异有统计学意义(P<0.05)。在肾组织中,黑面神组相对10%酒精处理组,硬脂酸含量升高(P<0.05)。结论 PLS-DA结果表明,黑面神诱导代谢扰动,而10%的酒精也可能诱发代谢扰动。
Objective Gas chromatography-mass spectrometry( GC-MS) coupled with metabolomic method was used to elucidate the effect of Breynia fruticosa( Euphorbiaceae),a folk medicine for the treatment of chronic bronchitis and inflammation by Dai in southern China,on liver and kidney of rats. Methods The rats were divided into the control group,Breynia fruticosa( 2 g/kg) treated group,10% alcohol treated group,and administrated continuously for 7 d. On the 8 thday,liver and kidney of the rats were collected from three groups and the extracts were derivatized and analyzed by GC-MS. Results According to the liver and kidney metabolomics results,Breynia fruticosa treated rats could be distinguished from control group and 10% alcohol treated group. In the liver samples,there is no difference between Breynia fruticosa treat group and control group,while the content of ribitol and D-glucose had significant difference between Breynia fruticosa treat group and 10% alcohol treated group( P 〈 0. 05). In the kidney samples,octadecanoic acid increased in Breynia fruticosa treated group compared to 10% alcohol treated group( P 〈 0. 05). Conclusion Partial least squares-discriminate analysis( PLS-DA) revealed that Breynia fruticosa induced metabolic perturbations,while the 10% alcohol could also induce metabolic perturbations.
