摘要 由于常规育种方式无法打破牡丹上胚轴休眠,导致牡丹育种周期较长,极大限制了牡丹生产的发展,所以寻求利用牡丹成熟胚组织培养技术对牡丹进行育种和快繁具有重要意义。本研究利用组织培养技术以‘凤丹白’成熟胚为材料研究了不同激素组合对胚生长、丛生芽诱导和生根的影响。研究发现培养基1/2MS+6-BA 1.5 mg/L+NAA 0.1 mg/L+GA31.0 mg/L对胚的萌发效果最好;培养基MS+6-BA 1.5 mg/L+NAA0.1 mg/L最有利于牡丹丛生芽诱导。研究表明,GA3有助于打破上胚轴休眠,利于种子萌动和茎的伸长生长,为进一步研究牡丹快速繁殖和育种提供了一些参考。 Since the conventional breeding method cannot break the dormancy of peony epicotyl, the breeding period of peony is longer and the development of peony production is limited greatly. Therefore, it is of great significance to seek the technology of tissue culture of mature embryo for breeding and rapid propagation of peony. In this study, we used tissue culture technology to study the effects of different hormonal combinations on the embryo growth, induction from embryonic shoots and rooting using the mature embryo of 'Fengdanbai'. The results showed that culture medium with 1/2 MS, 6-BA 1.5 mg/L, NAA 0.1 mg/L and GA3 1 mg/L had the best germination effect on embryos, and culture medium with MS, 6-BA 1.5 mg/L and NAA 0.1 mg/L was most conducive to the induction of clustered shoots of Paeonia s uffruticosa. The results showed that GA3 was helpful to break the dormancy of hypocotyl and germinate seeds, which might provide some references for further research on rapid propagation and breeding of peony.
