摘要
以桃儿七种子胚培养获得7 d生的无菌苗,切取无菌苗的下胚轴、子叶做为外植体,通过调控不同生长调节剂浓度配比,诱导培养获得愈伤组织,利用愈伤组织进行体细胞胚胎发生研究,观察体细胞胚胎的发育过程,分析不同激素配比对桃儿七体细胞胚发生的影响,从而建立桃儿七体细胞胚胎发生技术体系。本研究结果表明最适宜桃儿七愈伤组织诱导的培养基是MS+0.2 mg/L NAA+0.1 mg/L GA3+1.0 mg/L 6-BA,有利于胚性愈伤组织转化和增殖的培养基为MS+1.0 mg/L 6-BA+0.1 mg/L NAA+0.1 mg/L GA3+300 mg/L CH+40.0 g/L蔗糖,培养基MS+6-BA 0.5 mg/L+GA30.1 mg/L+NAA 0.01 mg/L有利于体细胞胚的增殖和萌发。
The callus was induced from hypocotyls and cotyledon of the 7 day old aseptic seedlings which were cultured with seed embryo of Sinopodophyllum emodi (Wall.) Ying by regulating the ratio of different growth regulators. The process of somatic embryogenesis and the effects on different ratio hormones were studied using the callus. Thereby establishing a somatic embryogenesis technology system ofSinopodophyllum emodi (Wall.) Ying. The results showed that the best medium was MS solid medium supplemented with 6-BA 1.0 mg/L and NAA 02 mg/L and GA3 0.1 mg/L. The suitable culture medium for the embryogenic calli transations and proliferation was M S + 1.0 mg/L 6-BA+0.1 mg/L NAA+0.1 mg/L GA3+300 mg/L CH+40.0 g/L sucrose. The optimal medium for somatic embryo development and germination was sold medium MS+6-BA 0.5 mg/L+GA30.1 mg/L+NAA 0.01 mg/L.
