摘要
许多蔷薇科(Rosaceae)委陵菜属(Potentilla L.)植物由于其含有多种次生代谢物,尤其是黄酮类物质,具有非常高的药用价值和经济价值.采用超声波震荡方法提取委陵菜属4种植物的总黄酮,通过单因素实验,确定了料液混和物质量浓度、乙醇含量、提取温度和提取时间这4个影响植物总黄酮提取的主要因素.在此基础上,设计了4因素3水平的正交试验,最终得到提取4种植物黄酮的最优方案.委陵菜在70℃,料液混合物10.0mg/mL,600mL/L乙醇,提取60min时效果最佳;皱叶委陵菜在70℃,料液混合物10.0mg/mL,700mL/L乙醇,提取50min时效果最佳;西山委陵菜在70℃,料液混合物10.0mg/mL,600mL/L乙醇,提取70min时效果最佳;翻白草在50℃,料液混合物10.0mg/mL,600mL/L乙醇,提取60min时效果最佳.这4种委陵菜属植物在它们各自相对应的提取最佳方案下得到的黄酮依次为13.126,34.712,36.482,45.973mg/g.实验流程简单,成本较低,毒性较小,适合于工业化的推广和应用.
Many plants in Rose family(Rosaceae)Potentillagenus(Potentilla L.)have high medicinal and economic value since they contain a variety of secondary metabolites,especially flavonoids.Single factor experiment was performed to determine the effect of four factors(including solid-liquid mass concentration,ethanol content,temperature and time)on the extraction of flavonoids from plants in Potentilla by using ultrasonic extraction.The conditions of flavonoids extraction from four plants were studied by using a 4-factor 3-level orthogonal test.For Potentilla,the best extraction effect was obtained under 70℃,using10.0 mg/mL solid-liquid and 600 mL/L ethanol,with 60 minutes’ extraction.For P.ancistrifolia,the condition for best extraction effect was 70 ℃,using 10.0 mg/mL solid-liquid and 700 mL/L ethanol,with 50 minutes’ extraction.For P.sischanensis,the optimal extraction condition was 70℃,using 10.0 mg/mL solid-liquid and 600 mL/L ethanol,with 70 minutes’ extraction.For P.discolor,the optimal extraction condition was 50 ℃,using 10.0 mg/mL solid-liquid and 600 mL/L ethanol,with 60 minutes’ extraction.The contents of total flavonoids extracted from the four plants under their optimal extraction conditions were13.126,34.712,36.482,45.973 mg/g,respectively.The extraction procedure that has been set up in this study is simple,cost-effective,less toxic,and is suitable for industrial application.
