摘要
目的观察RIN1基因(Ras interaction/interference1,RIN1)在创伤后应激障碍(PTSD)大鼠的表达变化,探讨RIN1基因参与调节创伤后应激障碍大鼠恐惧记忆异常的机制。方法采用国际认定的单一连续(SPS)刺激建立PTSD大鼠模型,应用条件恐惧实验检测大鼠恐惧记忆,采用免疫荧光共标和Western blot检测SPS大鼠海马和杏仁核中RIN1表达变化。结果条件性恐惧实验结果显示,在恐惧记忆形成阶段,SPS组大鼠与正常组大鼠相比Freezing时间明显增加。免疫荧光共标染色和Western blot检测显示,SPS组大鼠海马和杏仁核RIN1表达水平均增加。结论海马及杏仁核RIN1可能参与SPS大鼠恐惧记忆异常的调节。
Objective To observe the expression of Ras interaction/interference1(RIN1) gene in post-traumatic stress disorder(PTSD) rats, and to explore pathogenesis of PTSD rats. Methods The animal models of PTSD were conducted by single prolonged stress(SPS), the fear memory was detected by condition fear experiment, the changes of RIN1 expression in hippocampus and amygdala of SPS rats were detected by immunofluorescence and Western blot. Results At the stage of fear memory formation, freezing time in SPS group was significantly longer than that in normal group, the expression level of RIN1 mRNA and protein in hippocampus and amygdala was increased in SPS group than in normal control. Conclusion RIN1 in hippocampus and amygdala might be involved in the regulation of fear memory disorder in SPS rats.
