摘要
目的研究长链非编码RNA Taurine Up-Regulated 1(TUG1)基因调控膀胱尿路上皮癌(bladder urothelial carcinoma,BUC)细胞对化疗药物顺铂(CDDP)的敏感性。方法实时定量PCR法检测TUG1基因在CDDP耐药的BUC细胞T24/CDDP中的表达。RNA干扰沉默T24/CDDP细胞中TUG1的表达。增强型CCK8法检测T24/CDDP细胞的增殖活力以及对CDDP的敏感性。Annexin V-FITC/PI双染法检测T24/CDDP细胞的凋亡。结果与T24细胞相比,TUG1基因在T24/CDDP细胞中的表达显著上调。TUG1沉默能够提高T24/CDDP细胞对CDDP的敏感性,并增强CDDP诱导的细胞毒效应,即抑制T24/CDDP细胞的增殖活力,促进细胞凋亡。结论长链非编码RNA TUG1基因与BUC的化疗耐药相关,其表达沉默能够提高BUC细胞对CDDP的敏感性。
Objective To study the regulation of long noncoding RNA Taurine Up-Regulated 1(TUG1) gene on the sensitivity of bladder urothelial carcinoma(BUC) cells to cisplatin(CDDP). Methods Real-time quantitative PCR was used to detect the expression of TUG1 gene in the BUC T24/CDDP cells which resistant to CDDP. The expression of TUG1 was silenced by using RNA interference. The enhanced CCK8 assay was used to detect the proliferation of T24/CDDP cells and its sensitivity to CDDP. Annexin V-FITC/PI double staining assay was used to detect the apoptosis of T24/CDDP cells. Results Compared with T24 cells, the expression level of TUG1 gene in T24/CDDP cells was up-regulated significantly. TUG1 silencing improved the sensitivity of T24/CDDP cells to CDDP and promoted the cytotoxicity induced by CDDP, that is, significantly inhibited the proliferation and promoted cell apoptosis of T24/CDDP cells. Conclusion LncRNA TUG1 is related to chemotherapeutic resistance of BUC, and its expression silencing can increase the sensitivity of BUC cells to CDDP.
