摘要
目的研究miR-31在胃癌患者中的表达,探讨miR-31对胃癌HGC-27细胞增殖、侵袭和转移的影响。方法收集胃癌患者标本39例,同时收取相应的癌旁组织20例。通过荧光定量PCR方法检测各组织中miR-31的表达水平。以脂质体为载体将miR-31模拟物转染至HGC-27细胞,CCK-8方法检测细胞增殖能力变化,Transwell实验检测HGC-27细胞侵袭和迁移能力变化。结果 miR-31在胃癌组织中的表达水平明显低于癌旁组织,P<0.01。过表达miR-31能够显著抑制HGC-27细胞的增殖,显著抑制HGC-27细胞的侵袭和迁移能力(P<0.01)。结论 MiR-31在胃癌中低表达,并抑制胃癌细胞系HGC-27的增殖、迁移和侵袭。
Objective To investigate the expression of miR-31 in gastric cancer patients and explore the effect of miR-31 on the proliferation, invasion and metastasis of gastric cancer HGC-27 cells. Methods A total of 39 patients with gastric cancer was collected, while 20 corresponding para-cancerous tissues were collected. The expression level of miR-31 in tissues was detected by fluorescence quantitative PCR(qRT-PCR). The miR-31 mimics were transfected into HGC-27 cells with liposome, and cell proliferation was detected by CCK-8 method. The invasion and migration abiity of HGC-27 cells were detected by transwell test. Results The expression level of miR-31 in gastric cancer tissues was significantly lower than that in adjacent tissues(P〈0.01). The over-expression of miR-31 could significantly inhibit the proliferation, invasion and migration of HGC-27 cells. Conclusion MiR-31 is poorly expressed in gastric cancer and inhibits proliferation, migration and invasion of gastric cancer cell line HGC-27.
