高效液相色谱法测定拉萨市市场自产饮料合成色素

摘要 目的:正确评价高海拔地区高效液相色谱仪(HPLC)使用精确性的同时,建立HPLC测定拉萨市市售饮料中食用合成色素柠檬黄、苋菜红、胭脂红、日落黄含量的方法。方法:采用SHIMADZU VP-ODS C8色谱柱(5μm,4.6×150 L),以甲醇0.02 mol/L乙酸铵水溶液为流动相,梯度洗脱,流速为0.8 mL/min,柱温30℃,检测波长254 nm,进样量20μL。结果:柠檬黄、苋菜红、胭脂红、日落黄分别在100~3.125 mg/L(r=0.9999)、47~1.46875 mg/L(r=0.9996)、50~1.5625 mg/L(r=0.9994)、100~3.125 mg/L(r=0.9997)范围内呈良好的线性关系。结论:文章建立的食用合成色素高效液相检测方法准确、简便、可靠、灵敏度高、专属性好,可用于市售饮料中食用合成色素的质量控制。 A method of determining the content of edible synthetic pigments, such as tartrazine, amaranth, carmine, and sunset yellow, in commercially available beverages in Lhasa with HPLC was established and high-performance liquid chromatography (HPLC) using accurately in high-altitude areas was evaluated correctly. Chro- matographic analysis was achieved on a SHIMADZU VP-ODS C8 column (5 μm, 4.6 μm×150 ram) using methanol and 0.02 mol/L ammonium acetate solution as the flow phase for gradient elution with 0.8 mL/min flow volume at 30℃ column temperature, 254 nm of detection wavelength and 20 μL injection volume. The result showed that tartrazine, amaranth, carmine and sunset yellow had good linear relationship in the range of 100 ~ 3.125 mg/ L (r=0.9999), 47 ~ 1.46875 mg/L (r=0.9996), 50 ~ 1.5625 mg/L (r=0.9994), and 100 ~ 3.125 mg/L (r=0.9997) respectively. The HPLC method for the determination of edible synthetic pigments in beverages is accurate, simple, reliable, highly sensitive and specific. This method can be used for controlling the quality of edible pigments in commercial beverages.
