摘要
目的:探究牛磺酸对血管紧张素Ⅱ诱导的高血压是否有保护作用。方法:给小鼠皮下埋入血管紧张素Ⅱ微量泵诱导小鼠高血压模型,通过小鼠血压测定、主动脉HE染色、Masson染色等方法检测牛磺酸对高血压及其导致的血管重构作用;PCR探究牛磺酸对血管纤维化的作用;Western blot研究牛磺酸对血管紧张素Ⅱ诱导的高血压性血管重构的分子机制的影响。结果:牛磺酸抑制血管紧张素Ⅱ诱导的高血压及由此产生的血管重构,抑制m RNA水平纤维化标志物的表达,抑制血管紧张素Ⅱ作用下MAPK通路的激活。结论:牛磺酸可能通过抑制MAPK通路的激活对血管紧张素Ⅱ引起的高血压及由此产生的血管重构发挥保护作用。
Objective:To investigate if taurine has the protective effect on angiotensin Ⅱ-induced hypertension. Methods:AngiotensinⅡosmotic mini-pumps were implanted to mice subcutaneously to induce hypertension. Caudal arterial pressure was tested,and Hematoxylin-eosin staining and Masson staining were performed to determine the effect of taurine on hypertension and hypertension induced vascular remodelling;the m RNA levels of fibrotic markers were determined by RT-PCR;Western blot was performed to investigate the effect of taurine on molecular mechanisms of angiotensin Ⅱ-induced hypertension and hypertension induced vascular remodelling. Results:Taurine inhibited angiotensinⅡ-induced hypertension and consequent vascular remodelling,reduced the m RNA levels of fibrotic markers,and inhibited the MAPK pathway during the process. Conclusion:Taurine cowd inhibit MAPK pathway to attenuate angiotensinⅡ-induced hypertension and consequent vascular remodelling.
