摘要
目的:建立一种可快速检测大鼠血浆中舒芬太尼浓度的薄层色谱-表面增强拉曼光谱法(thin layer chromatography-surface enhanced Raman spectroscopy,TLC-SERS)。方法:以二氯甲烷-甲醇(9∶1.2)为展开剂,采用TLC法对血浆样品进行分离,然后用SERS法对舒芬太尼进行定性、定量分析。将舒芬太尼的浓度与拉曼位移1004cm-1处的特征峰强度做线性回归,考察该方法的标准曲线、检测限和回收率。结果:舒芬太尼在0.85~85.00μg/ml浓度范围内与SERS在1004cm-1处的峰强度呈良好线性关系,检测限为0.85μg/ml,低浓度(4.25μg/ml)和高浓度(42.50μg/ml)的回收率分别为(86.00±8.15)%和(103.45±3.59)%(n=3)。结论:本研究为血浆中舒芬太尼的浓度检测提供了一种便捷、快速、低成本的检测方法。
Objective : To establish a method for rapid determination of sufentanil concentration in rat plasma by thin layer chromatography surface enhanced Raman spectroscopy (TLC SERS) method .Methods: The plasma samples were separated by TLC with diehloromethane-methanol(9:1.2) as a developing agent ,and then SERS ,was applied for qualitative and quantitative analysis of sufentanil .Linear regression was performed between the concentration of sufentanil and the intensity of the Raman characteristic peak at 1004 em 1 .The standard curve ,detection limit and reeovery rate were all investigated .Results : The con eentration of sufentanil within the range of 0.85 85.00/zg/ml and the peak intensity of SERS at 1004 em 1 were in good lineari ty .The detection limit was 0.85 μg/ml,and the recovery rates of low (4. 25 μg/ml) and high concentrations (42.50 μg/ml ) were (86.00±8.15)% and (103.45±3.59)% (n=3 ) ,respectively .Conclusion : The study established a simple ,rapid and prac tical method for the determination of sufentanil plasma concentration .
