摘要
目的分析高突足襞蛞蝓提取物多糖特征并对其抗氧化活性。方法采用水提、Sevage法除蛋白得到受试物;气相色谱-质谱(GC-MS)、高效液相色谱(HPLC)分析受试物单糖组成,凝胶渗透色谱(GPC)分析相对分子质量及其分布; CSE暴露诱导气道上皮细胞氧化应激观察受试物抗氧化效果。结果高突足襞蛞蝓提取物中多糖是由核糖、鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖组成,经脱蛋白处理后单糖组成不变,质量分数有所差别;受试物多糖数均相对分子质量(Mn)为1. 33×10~4,重均分子量(Mw)为3. 90×10~5;脱蛋白受试物多糖M n为4. 57×10~3,Mw为1. 40×10~4;受试物能降低烟草烟雾提取物(CSE)诱导的气道上皮细胞ROS生成,增加SOD酶活力(P <0. 01)。结论蛞蝓提取物含有多种单糖组成的杂多糖,且具有显著抗CSE诱导的气道上皮氧化应激作用。
Objective To study the characteristics and antioxidant activities of polysaccharides from the extracts of Vaginulus alte Ferussac. Methods The mixed polysaccharide extracts were acquired by successively removing the protein components using water extraction and Sevage method. Subsequently, Gas chromatography - mass spectrometry ( GC - MS), high performance liquid chromatography (HPLC) were performed to analyze the composition of the extracts; gel permeation chromatography (GPC) was used to analyze the relative molecular weight and their distribution. Finally, the antioxidant effects were studied by observing the inhibitory activity to oxidative stress induced by cigarette smoke extracts (CSE) in airway epithelial cells. Results The polysaccharides in the extracts of Vaginulus alte Ferussac were mainly composed by ribose, rhamnose, arabinose, xylose, mannose, glucose and galactose, and the monosaccharide composition remains unchanged after deproteinizing treatment. The numberaverage molecular weight (Mn) of the polysaccharide extracts is 1.33 x 104, the weight - average molecular weight (Mw) is 3.90 × 10 5 ; the Mn of the deproteinized extracts is 4.57 × 10 3, Mw is 1.40× 10 4. The polysaccharide extracts can significantly reduce reactive oxygen species (ROS) production and increase Superoxide Dismutase (SOD) activity in CSE inducing airway epithelial cells. Conclusion the extracts from Vaginulus alte Ferussac contain various heteropolysaccharides composed by distinctive monosaccharides, and the extracts can significantly inhibit the oxidative stress induced by CSE in airway epithelial cells.
