决枳合剂治疗酒精性脂肪肝大鼠的实验研究 被引量:3

Experimental study of the mixture of cassia seed and hovenia dulcis thunb in the treatment of rats with alcoholic fatty liver

摘要 目的研究决枳合剂对酒精性脂肪肝大鼠肝组织病理、肝功能和炎症因子的影响。方法用56%酒精灌胃建立酒精性脂肪肝模型,并随机分为模型组、对照A组、对照B组和实验组。另取正常大鼠设为空白组。空白组和模型组均予以10 mL·kg^(-1)0.9%Na Cl,qd;对照A组按10 mL·kg^(-1)剂量予以1.5g·mL^(-1)决明子,qd;对照B组按10 mL·kg^(-1)剂量予以1.5 g·mL^(-1)枳椇子,qd;实验组按10 mL·kg^(-1)剂量予以1.5 g·mL^(-1)决枳合剂,qd。5组大鼠均灌胃给药4周。比较5组大鼠的肝组织病理结果、谷丙转氨酶(ALT)、谷草转氨酶(AST)、核转录因子-κB(NF-κB)、肿瘤坏死因子(TNF-α)和白细胞介素-6(IL-6)水平。结果空白组、模型组、对照A组、对照B组和实验组的肝炎活动度评分分别为(0.57±0.48),(3.71±0.85),(2.52±0.79),(2.37±0.81)和(1.61±0.67)分,ALT分别为(39.25±10.07),(89.37±14.53),(72.43±12.08),(70.17±13.56)和(51.26±11.57)U·L^(-1),AST分别为(34.14±4.95),(139.19±14.87),(90.00±17.57),(87.47±12.38)和(51.27±8.25)U·L^(-1),NF-κB分别为(265.45±47.12),(534.41±72.18),(387.15±60.29),(390.31±63.64)和(321.26±43.25)ng·L^(-1),TNF-α分别为(26.85±4.97),(89.78±9.86),(55.21±11.79),(57.32±9.63)和(40.24±9.47)nmol·L^(-1),IL-6分别为(12.13±5.86),(69.34±12.79),(43.16±10.62),(46.28±11.57)和(24.47±10.84)pg·mL^(-1),实验组的上述指标与其他4组比较,差异均有统计学意义(均P<0.05)。结论决枳合剂能够明显改善酒精性脂肪肝大鼠肝组织的病理变化和肝功能,且能够下调炎症因子的表达。 Objective To explore the effects of the mixture of cassia seed and hovenia dulcis thunb on liver pathology,liver function and inflammatory factors of rats with alcoholic fatty liver.Methods Alcoholic fatty liver model was established by intragastric administration of 56% alcohol,and the model rats were randomly divided into model group,control A group,control B group and test group.And the normal rats were set up as blank group.Blank group and model group were treated with 10mL·kg^-10.9% NaCl,qd.Control A group was given 1.5 g·mL^-1cassia seed at the dose of 10 mL·kg^-1,qd.Control B group was given 1.5 g·mL^-1hovenia dulcis thunb at the dose of 10 mL·kg^-1,qd.Test group was given 1.5 g·mL^-1mixture of cassia seed and hovenia dulcis thunb at the dose of 10 mL·kg^-1,qd.Five groups were administrated by gavage for 4 weeks.The liver pathological results,alanine ami-notransferase(ALT),aspartate aminotransferase(AST),nuclear factor-kappa B(NF-κB),tumor necrosis factor(TNF-α)and interleukin-6(IL-6)were compared between the five groups.ResultsThe main indexes of blank,model,control A,control B and test groups were compared:liver inflammation activity score were(0.57±0.48),(3.71±0.85),(2.52±0.79),(2.37±0.81)and(1.61±0.67),ALT were(39.25±10.07),(89.37±14.53),(72.43±12.08),(70.17±13.56)and(51.26±11.57)U·L^-1,AST were(34.14±4.95),(139.19±14.87),(90.00±17.57),(87.47±12.38)and(51.27±8.25)U·L^-1,NF-κB were(265.45±47.12),(534.41±72.18),(387.15±60.29),(390.31±63.64)and(321.26±43.25)ng·L^-1,TNF-αwere(26.85±4.97),(89.78±9.86),(55.21±11.79),(57.32±9.63)and(40.24±9.47)nmol·L^-1,IL-6 were(12.13±5.86),(69.34±12.79),(43.16±10.62),(46.28±11.57)and(24.47±10.84)pg·mL^-1,the differences were statistically significant between test group and other four groups(all P〈0.05).Conclusion The mixture of cassia seed and hovenia dulcis thunb could obviously improve the pathological changes of liver tissue and liver function,and also could reduce the expression of inflammatory factors in rats with alcoholic fatty liver disease.
