α-细辛脑亚微乳对阿尔茨海默症大鼠空间学习记忆能力的改善作用 被引量:3

Improvement of α-asarone submicron emulsion on the spatial learning and memory ability of rats with Alzheimer’s disease

摘要 目的研究α-细辛脑亚微乳对阿尔茨海默症(AD)大鼠空间学习记忆能力的改善作用。方法给予SD大鼠三氯化铝(20 mg·kg(-1),ig)联合D-半乳糖(180 mg·kg(-1),ip)建立AD模型,将造模成功的大鼠随机分为细辛脑亚微乳低、高剂量(10、25 mg·kg(-1))组、盐酸多奈哌齐(1. 75 mg·kg(-1))组及模型组,每日给药1次,连续28 d;采用Morris水迷宫空间探索法测定大鼠给药前、后穿越平台的次数;采用HE染色法观察大鼠海马区的组织形态;采用免疫组化染色法检测β-淀粉样蛋白(Aβ1-42)、Tau蛋白的表达水平;测定大鼠脑内乙酰胆碱(ACh)、乙酰胆碱酯酶、胆碱乙酰转移酶的含量。结果与模型组比较,α-细辛脑亚微乳高剂量可显著增加AD大鼠穿越平台的次数,其作用可能与减少大鼠海马区神经元的缺失,降低Aβ、Tau蛋白的表达水平,增加脑组织中ACh的含量有关。结论α-细辛脑亚微乳可显著改善AD大鼠的认知缺陷与记忆障碍,具有治疗AD的潜在应用前景。 OBJECTIVE To study the effect of α-asarone submicron emulsion( ASA-SE) on the spatial learning and memory ability of rats with Alzheimer's disease( AD). METHODS The SD rats were administrated aluminum chloride(20 mg·kg(-1),ip) and D-galactose(180 mg·kg(-1),ig)to establish the AD model. The AD rats were randomly divided into four groups such as 10 mg·kg(-1) ASA-SE group,25 mg·kg(-1) ASA-SE group,Donepezil hydrochloride(1. 75 mg·kg(-1)) group and model group. All rats in each group were dosed once a day as planned and 28 days in total. The Morris water maze was applied to evaluate the changes of the across platform times before and after treatment. The neuron morphology was observed with HE staining. The β-amyloid( Aβ1-42)and tau protein in hippocampus were imaged by Immunohistochemical staining method. Besides,the content of acetylcholine( ACh),acetylcholinesterase and choline acetyltransferase in the brains of rats were measured. RESULTS Compared with the model group,25 mg·kg(-1) ASA-SE can significantly increase the platform crossing time in the AD rats,which might be attributed to the ameliorated neuron loss,Aβ and tau protein level and the increased content of ACh. CONCLUSION The ASA-SE demonstrates certain efficacy in improving the cognitive deficits and memory impairment,which provides a promising therapy for treating AD.
