摘要
目的:基于转录组测序技术探索风湿宁胶囊(FSN)治疗RA的关键基因。方法:大鼠按随机数字表法分为正常、CIA、复合模型、FSN-CIA低、中、高及FSN-复合低、中、高、Tofacitinib、塞隆风湿组各12只,正常组、模型组生理盐水灌胃,给药组FSN每日1次28 d,通过关节肿胀度、关节病理、相关炎性细胞水平观察FSN治疗效果;Illumina Hi Seq 4000测序分析正常、CIA、复合模型、FSN-CIA高剂量、FSN-复合高剂量5组大鼠,对差异基因进行GO、KEGG分析。结果:模型组大鼠关节肿胀度、炎性细胞因子水平明显升高,治疗组关节红肿减轻、关节病理特征改善,炎性细胞因子水平降低,共检测9070个新转录本,GO分析表明,差异基因富集在细胞进程、代谢、免疫等;KEGG分析表明,差异基因富集在遗传信息进程、人类疾病等,复合模型与其给药组、CIA与复合模型组、CIA与其给药组差异基因分别上调54、1和65个以及下调30、2和0个。结论:FSN胶囊可明显改善CIA大鼠相关生理指标及关节病理特征,YB-1可能为FSN胶囊治疗RA的关键蛋白之一。
Objective: To explore the key differentially expressed genes of rheumatoid arthritis based on transcriptome sequencing( RNA-seq). Methods: Rats were randomly divided into normal group,CIA group,composite model group,FSN low,medium and high dose groups,each with 12 rats. The normal group and the model group were intragastrically administrated with the same amount of normal saline. The FSN of each dose group was once a day for 28 days. After 28 days,the degree of joint swelling,joint pathology,and inflammatory cytokine levels were observed. The therapeutic effects of FSN capsules on different models of rats were observed. Five groups of rats in normal group,CIA model group,CIA +cold-wet group( compound model),FSN-CIA group and FSN-compound group were analyzed by Illumina Hi Seq 4000 sequencing platform,and the differentially expressed genes were analyzed by GO and KEGG pathway. Results: A total of9070 new transcripts were detected. GO enrichment analysis showed that differentially expressed genes were enriched in cellular processes,metabolism and immunity. KEGG pathway analysis showed that most of the differentially expressed genes were enriched in the process of genetic information,human diseases,and so on. The differentially expressed genes in CIA model group,CIA model group and CIA model group and CIA model group were up-regulated by 54,1 and 65,and down-regulated by 30,2 and 0. Conclusion: YB-1 may be one of the key proteins in the treatment of RA with FSN capsule.
