摘要
目的:观察针灸对骨髓抑制小鼠细胞DNA碱基切除修复基因XRCC1、ADPRT的调控作用。方法:选用清洁级、雄性昆明小鼠40只,体质量(20±2)g,自由饲养3 d后按体质量随机分为空白组、模型组、针刺组、艾灸组,并用CTX造成骨髓抑制模型。针刺组、艾灸组分别选取"大椎""膈俞""肾俞""足三里"施以针刺、艾灸治疗,空白组、模型组每日陪同固定不予治疗,每日1次,共治疗5 d,于第6天取各组小鼠双侧股骨,用ELISA法检测小鼠骨髓细胞DNA中XRCC1、ADPRT的含量变化。结果:与空白组比较,模型组、针刺组和艾灸组XRCC1、ADPRT含量均降低;与模型组比较,针刺组、艾灸组XRCC1、ADPRT含量均升高,表明针刺和艾灸能够显著上调CTX化疗小鼠骨髓细胞DNA中XRCC1、ADPRT的表达含量,提高DNA碱基切除修复能力,减轻骨髓抑制。结论:通过针刺、艾灸促进骨髓细胞DNA碱基切除修复能力是针灸改善骨髓抑制的关键机制之一。
Objective: To observe the effects of acupuncture and moxibustion on regulation of DNA base excision repair genes XRCC1 and ADPRT in the bone inhibition's mice. Methods: Forty clean-grade weight 20 ± 2 grams male Kunming mice were selected and randomly divided into a blank group,a model group,an acupuncture group and a moxibustion group after free for 3 days. With cyclophospha-mide( CTX) to establish mice models of marrow inhibition. Mice in the acupuncture group and moxibustion group were treated with acupuncture or moxibustion at"Du-zhui "( GV14),"Geshu "( BL17),"Shenshu"( BL23) and "Zusanli"( ST36),respectively. Mice in the blank group and model group were immobilized without any treatment. All the treatments were given once a day for consecutive 5 days. On the sixth day all the mice femur were collected. The ELISA method was applied to measure the contents change of XRCC1 and ADPRT in marrow cells of all the mice. Results: Compared with the blank group,the contents of XRCC1 and ADPRT in marrow cells in the model group,an acupuncture group and a moxibustion group were reduced; compared with the model group,the contents of XRCC1 and ADPRT in marrow cells in the acupuncture group and moxibustion group were increased. Acupuncture and moxibustion markedly up-regulated the contents of XRCC1 and ADPRT in marrow cells of Cyclophosphamide-induced mice,which could improve the DNA base excision repair ability and relieve the myelosuppression. Conclusion: For Acupuncture and moxibustion,one of the key mechanisms of acupuncture and moxibustion in the treatment of bone marrow suppression is that it can promote the DNA base escision repair ability.
