不同产地黄连饮片标准汤剂制备与质量标准研究

Preparation and Quality Standard Study on the Coptis Chinensis Standard Decoction from Different Areas

目的:制备黄连饮片标准汤剂,并对其进行质量标准研究。方法:以水为溶媒,采用标准化工艺制备标准汤剂,计算出膏率,采用HPLC法测定表小檗碱、黄连碱、巴马汀、小檗碱含量,并计算各成分含量转移率。结果:15批黄连标准汤剂的出膏率范围为17.53%～26.82%,均值为20.36%,标准偏差为3.34%;表小檗碱含量范围为36.51～48.05mg/g,均值为44.21mg/g,标准偏差为4.15mg/g,其转移率范围为55.59%～70.72%,均值为62.80%,标准偏差为4.90%;黄连碱含量范围为42.89～54.68mg/g,均值为50.89mg/g,标准偏差为4.18mg/g,其转移率范围为52.50%～64.86%,均值为57.76%,标准偏差为3.70%;巴马汀含量范围为41.30～46.78mg/g,均值为44.08mg/g,标准偏差为1.44mg/g,其转移率范围为56.42%～67.94%,均值为61.48%,标准偏差为4.00%;小檗碱含量范围为149.8～185.6mg/g,均值为169.18mg/g,标准偏差为11.35mg/g,其转移率范围为53.32%～65.97%,均值为59.68%,标准偏差为3.65%;指纹图谱得到共有峰9个,相似度在0.99以上,黄连饮片与标准汤剂指纹图谱具有较好的一致性。结论:15批不同产地黄连标准汤剂与其饮片的基本属性一致,本研究黄连饮片标准汤剂制备方法规范,可用于黄连标准汤剂的制备及质量标准研究。

Objective ： To prepare the standard decoction and study the quality standards of Coptis Chinensis. Methods. The standard decoction of Coptis Chinensis was preparated with water by using standardization method, and calculated the extractum rate of the standard decoction. The content of epiberberine, coptisine, palmatine and berberine was determined by HPLC, and calculated the transfer rate of the standard decoction. Results： The rate of extractum in 15 batches of standard decoction was 17.53%-26.82%, the average yield was 20.36% and the standard deviation was 3.34%. The content of epiberberine was 36.51-48.05mg/g, the mean value was 44.21 mg/g and the standard deviation was 4.15mg/g. The transfer rate of epiberberine was 55.59-70.72mg/g, the mean value was 62.80% and the standard deviation was 4.90%. The content of coptisine was 42.89-54.68mg/g,the mean value was 50.89 mg/g and the standard deviation was 4. 18mg/g. The transfer rate of coptisine was 52.50-64.86mg/g,the mean value was 57.76% and the standard deviation was 3.70%. The content of palmatine was 41.30-46.78mg/g,the mean value was 44.08 mg/g and the standard deviation was 1.44mg/g. The transfer rate of palmatine was 56. 42-67. 94mg/g,the mean value was 61. 48% and the standard deviation was 4.00%. The content of berberine was 149.8-185. 6mg/g,the mean value was 169. 18 mg/g and the standard deviation was 11.35 mg/g. The transfer rate of berberine was 53. 32-65.97mg/g,the mean value was 59. 68% and the standard deviation was 3.65 %. There were nine common peaks, and the similarity was more than 0.99, which created a consistent connection between the 15 batches of Coptis Chinensis standard decoction and their pieces. Conclusion. The basic properties of 15 batches of Coptis Chinensis standard decoction from different areas are consistent with their pieces. It is demonstrated that the method for preparing the standard decoction is normative, which can be used to the preparation and quality standard studies of the standard decoction of Coptis Chinensis.
