摘要
目的观察褪黑素对兔耳增生性瘢痕Ⅰ、Ⅲ型胶原的影响。方法建立兔耳增生性瘢痕动物模型,设褪黑素实验组及空白对照组。实验组每天腹腔内注射5.0mg/kg褪黑激素;空白对照组每天腹腔内注射等量生理盐水。饲养6周后,收集瘢痕组织、固定标本。苏木素-伊红(HE)染色组织测量瘢痕厚度、天狼星红染色测量胶原所占面积百分比。结果第42天时观察实验组瘢痕增生程度明显低于对照组,实验组瘢痕组织厚度为(920.30±227.56)μm,对照组厚度为(1033.10±263.59)μm,对照组瘢痕增生程度强于实验组(P=0.000)。实验组Ⅰ型胶原含量(0.115±0.051)%,对照组Ⅰ型胶原含量为(0.148±0.043)%,实验组I型胶原含量低于对照组(P=0.026);Ⅲ型胶原含量实验组为(0.145±0.041)%,对照组含量为(0.121±0.034)%。实验组Ⅲ型胶原含量高于对照组(P=0.013)。结论褪黑素可以通过调节Ⅰ、Ⅲ型胶原含量比例,从而减轻瘢痕增生。
Objective To observe the effect of melatonin on the expression of type I collagen and type Ⅲ collagen in hypertrophic scar of rabbit ears. Methods The animal model of hypertrophic sear of rabbit ears was established, and the experimental group of melatonin and the blank control group were set up. Experimental group : intraperitoneal injection of 5.0 mg/kg melatonin every day ; blank control group - intraperitoneal injection of equal amount of saline daily. After 6 weeks of rearing, cieatrieial tissue and fixed specimen were collected. The thickness of the scar was measured by hematoxylin- eosin (HE) stai- ning and the percentage of the area of collagen was measured by Sirius red staining. Results On the forty - second day, the degree of scar hyperplasia in the experimental group was significantly lower than that in the control group. The thickness of scar tissue in the experimental group was (920. 30-± 227. 56) μm, and the thickness of the control group was ( 1 033.10 ± 263.59) μm. The scar hyperplasia in the control group was stronger than that in the experimental group ( P = 0. 000). The content of type I collagen in experimental group was (0. 115 ±0.051 )%, and the content of type I collagen in control group was (0. 148 ± 0. 043 ) % , the content of type [ collagen in the experimental group was lower than that of the control group (P =0.026 ). The content of type Ill collagen in experimental group was (0. 145 ± 0. 041 ) %, while that in control group was (0. 121 ± 0. 034) %, the content of type Ill collagen in the experimental group was higher than that of the control group (P = 0. 013 ). Conclusion The results indicate that melatonin can reduce scar proliferation through regulating the proportion of collagen type I and type III.
