The expression of adenosine di - phosphate.ribosylation factor guanosine triphosph - ase activatingprotein 1 in papillary thyroid carcinoma

摘要 目的观察二磷酸腺苷(ADP)核糖化因子鸟苷酸激酶1(ASAPl)在甲状腺乳头状癌发生发展中的作用。方法采用免疫组织荧光、实时定量聚合酶链反应(Real-timePCR)及Westernblot检测ASAPl在28例甲状腺乳头状癌及癌旁组织中的表达。结果甲状腺乳头状癌和癌旁组织中ASAPl蛋白荧光信号主要定位于细胞质,甲状腺乳头状癌组织中ASAPl蛋白的平均荧光值中位数为202.50,癌旁组织中ASAP1蛋白平均荧光值中位数为72.50,两组差异有统计学意义(P=0.000);ASAP1mRNA在甲状腺乳头状癌组织中的相对表达量(0.802±0.126)显著高于癌旁组织(0.142±0.023)且差异有统计学意义(P=0.000);甲状腺乳头状癌组织中ASAPl蛋白的相对表达量(0.697±0.138)高于癌旁组织(0.129±0.037)且差异有统计学意义(P=0.000)。结论ASAPl在甲状腺乳头状癌组织中呈明显过表达,在甲状腺乳头状癌的发生发展中可能起重要作用。 Objective To explore the expression and significance of adenosine di - phosphate (ADP) - ribosylation factor guanosine tfiphosph (GTP) - ase activating protein 1 (ASAP1) protein in papillary thyroid carcinoma. Methods The expression levels of ASAP1 in 28 cases of papillary thyroid carcinoma tissues and adjacent normal thyroid tissues were detected by immunofluorescence, real -time quantitative polymerase chain reaction (Real- time PCR) and Western blotting. Results ASAP1 fluorescence signal in papillary thyroid carcinoma and adjacent tissues was mainly located in cytoplasm, the median fluo- rescence value of ASAP1 protein in thyroid papillary carcinoma was 202. 50, and the median fluorescence value of adjacent tissues was 72. 50, the difference between the two groups was statistically significant (P = 0. 000 ) ; the relative expression of ASAP1 mRNA (0. 802 ± 0. 126 ) in papillary thyroid careinoma was significantly higher than that in adjacent normal thyroid tissues ( 0. 142 ± 0. 023 ) and the difference was statistically significant (P =0. 000) ; the relative expression of ASAP1 protein was (0. 697 ±0. 138) in thyroid papillary carcinoma, which were over -expressed comparing with adjacent normal thyroid tissues (0. 129 ± 0. 037 ) and the difference was statistically signifieant ( P = 0. 000 ). Conclusion Over - expression of ASAP1 might play a key role in the papillary thyroid carcinoma.
