摘要
目的优化杜仲籽中桃叶珊瑚苷的提取工艺。方法以提取液中桃叶珊瑚苷浓度为响应值,在单因素考察的基础上,采用Box-Behnken Design响应面法,对甲醇体积分数、提取时间、料液比3个因素进行优化。结果优化后的提取条件:甲醇体积分数100%,提取时间40 min,液料比9.43 m L/g,该条件下桃叶珊瑚苷的提取浓度达到最大值(1.45 mg/m L),与理论值基本符合。将桃叶珊瑚苷的提取率从优化前的9.6 mg/g提高到29.0 mg/g,提取率提高了2倍。结论响应面法可用于优化杜仲籽中桃叶珊瑚苷的提取工艺。
Objective To optimize the conditions for extraction of aucubin from seeds of Eucommia ulmoides Oliver. Methods The effects of methanol concentration,extraction time and ratio of material to solution on the yield of aucubin were investigated. The extraction parameters were optimized with response surface methodology on the basis of single factor experiments using a three-factor and three-level Box-Behnken center united design. Results The optimal conditions of extraction were as follows,100% aqueous methanol solution as extraction solvent,extracting for 40 min and liquid/solid ratio of 9.43 mL/g. Under the optimized conditions,the concentration of aucubin of the extract was 1.45 mg/mL. The yield of aucubin from seeds of E. ulmoides increased by 2 times,from 9.6 mg/g to 29.0 mg/g. Conclusion Design-response surface methodology could efficiently predict the optimum extraction process of aucubin from seeds of E. ulmoides .
