摘要
目的研究青藤碱对人肝癌细胞HepG2的抑制作用。方法采用CCK-8法、流式细胞术分别检测青藤碱对人肝癌细胞HepG2增殖、凋亡和细胞周期的影响。结果 0.3、0.6、1.2、2.4、4.8 mmol/L的青藤碱可以抑制人肝癌细胞HepG2的增殖,并且随着浓度升高,抑制作用增强。0.3、0.6、1.2 mmol/L的青藤碱作用于人肝癌细胞HepG2 24 h可以使细胞周期阻滞在G1期,使进入S期的细胞比例降低,并且HepG2细胞凋亡显著增加。结论青藤碱可以有效抑制人肝癌细胞HepG2的增殖,诱导其凋亡,发挥抗肿瘤作用。
Objective To investigate the inhibitory effect of sinomenine on human hepatocellular carcinoma HepG2 cells. Methods CCK-8 and flow cytometry were used to check the proliferation,cell cycle and apoptosis of HepG2 cells after treatment with sinomenine. Results Sinomenine at different concentrations (0.3,0.6,1.2,2.4 and 4.8 mmol/L) inhibited the proliferation of HepG2 cells,and the inhibition effect was increased in a dose-dependent manner. Sinomenine (0.3,0.6,and 1.2 mmol/L) arrested the cell cycle of HepG2 cells at G1 phase,reduced the proportion of cells entering S phase and promoted HepG2 cell apoptosis at 24 h. Conclusion Sinomenine can effectively inhibit the proliferation of HepG2 cells and promote cell apoptosis.
