原儿茶酸、白杨素对非酒精性脂肪肝细胞模型的抗氧化作用 被引量:17

摘要 目的:探讨原儿茶酸、白杨素对非酒精性脂肪肝细胞模型的抗氧化应激作用。方法:人肝细胞L02,随机分为对照组、模型组、原儿茶酸组、白杨素组。除对照组外,其余3组细胞均以20%脂肪乳诱导非酒精性脂肪肝细胞模型。模型诱导成功后,原儿茶酸组添加1. 0μmol/L原儿茶酸,白杨素组添加20. 0μmol/L白杨素,培养24h。收集细胞上清液检测AST、ALT的含量;收集细胞裂解液检测MDA、GSH-Px、TG含量及SOD的活性。结果:(1)与对照组比较,模型组的AST、ALT、TG、MDA水平明显升高,差异有统计学意义(P <0. 05); GSH-Px和SOD的含量明显下降,差异有统计学意义(P <0. 05)。(2)与模型组比较,原儿茶酸组及白杨素组的AST、ALT、TG、MDA水平明显下降,差异有统计学意义(P <0. 05); GSH-Px和SOD的含量明显升高,差异有统计学意义(P <0.05)。(3)与原儿茶酸组比较,白杨素组的AST、ALT、TG、MDA水平变化不明显,差异无统计学意义(P> 0. 05); GSH-Px和SOD的含量变化不明显,差异无统计学意义(P> 0. 05)。结论:原儿茶酸、白杨素可通过抗氧化应激达到保护非酒精性脂肪肝细胞的作用。 Objective: To investigate the Anti-antioxidant role of Protocatechuic Acid and chrysin on Non-alcoholic fatty liver cells. Methods: Hu- man liver cells LO2 were divided into control group, model group, Protocatechuic Acid group and chrysin group. Except the control group, all the other groups were added 20% fat emulsion to establish Non-alcoholic fatty liver cell model. After the cell model were established, 1. 0 mol/L Protocatechuic Acid were added to the Protocatechuic Acid group, and 20.0 mol/L chrysin were added to the chrysin group. The cells were cultivate for 24 hours. The cells supernatant was collected to detected the levels of AST and ALT. Pyrolysis liquid was collected to detected the levels of MDA, GSH-Px, TG and SOD. Results:l. Compared with the control group, the levels of AST, ALT, TG and MDA in the model group were significantly increased, the difference was statistically significant. While the levels of GSH-Px and SOD were signifi- cantly decreased, the difference was statistically significant. 2. Compared with the model group, the levels of AST, ALT, TG and MDA in Protocatechuic Acid group and chrysin group were significantly decreased, the difference was statistically significant. While the levels of GSH-Px and SOD were significantly increased, the difference was statistically significant. 3. Compared with Protocatechuic Acid group, the levels of AST, ALT, TG, MDA, GSH-Px and SOD in chrysin group were no significant decreased. Conclusion:Protocatechuic Acid and chry- sin can protect Non-alcoholic fatty liver cells by inhibiting the oxidative stress.
