布地奈德雾化吸入对支气管哮喘小鼠肺组织NF-κB及OSM表达的影响 被引量:3

摘要 目的探讨布地奈德雾化吸入对支气管哮喘小鼠肺组织核因子-k B(NF-κB)及抑瘤素M(OSM)表达的影响。方法选取30只6-8周龄健康SPF级BALB/c小鼠作为本次研究对象,按随机数表法分为三组,即对照组、哮喘组(哮喘模型)、BUD组(哮喘模型行布地奈德干预),每组各10只。对照组采用生理盐水进行致敏和激发,哮喘模型建立采用腹腔注射卵清蛋白和氢氧化铝致敏以及雾化吸入卵清蛋白溶液激发哮喘,BUD组在致敏后BUD每次激发前雾化吸入布地奈德。于末次激发后24h内,取小鼠肺组织行RT-PCR及免疫组化,检测三组小鼠肺组织NF-ΚB和OSM mRNA和蛋白的表达水平。结果哮喘组和BUD组NF-κB、OSM阳性细胞百分比以及NF-κB、OSM mRNA水平均高于对照组,而BUD组上述指标均较哮喘组降低,组间比较差异具有统计学意义(P <0. 05)。结论支气管哮喘小鼠肺组织NF-κB及OSM表达增加,这可能是导致哮喘发生的机制之一。BUD可通过抑制NF-κB及OSM的表达水平而起到治疗哮喘的作用。 Objective To investigate the effect of inhaled budesonide on the expression of nuclear factor-kappaB (NF-κB) and oncostatin M (OSM) in bronchoalveolar lavage mice. Methods 30 healthy SPF BALB/ c mice aged 6 to 8 weeks were selected as the subjects and divided into three groups according to random number table: control group, asthma group (asthma model), BUD group (asthma model group Budesonide intervention), each group of 10. The control group was sensitized and stimulated with saline. The asthma model was established by intraperitoneal injection of ovalbumin and aluminum hydroxide sensitization and inhalation of ovalbumin solution to stimulate asthma. The BUD group was inhaled before each challenge of BUD after budesonide sensitization. 24 hours after the last challenge, the expression of NF-κB and OSM mRNA and protein in the lungs of the three groups were detected by RT-PCR and immunohistochemistry. Results The percentages of NF-κB and OSM positive cells and the levels of NF-κB and OSM mRNA in the asthma group and the BUD group were higher than those in the control group, while those in the BUD group were lower than those in the asthma group ( P〈 0.001). Conclusion The increased expression of NF- κB and OSM in the lung tissue of asthmatic mice may be one of the mechanisms leading to asthma. BUD may play a role in the treatment of asthma by inhibiting the expression of NF-κ B and OSM.
