中国甘蔗主要杂交亲本病毒性病害的分子鉴定 被引量:9

Molecular Identification of Viral Diseases in Major Sugarcane Hybrid Parents in China

摘要 了解当前中国甘蔗主要杂交亲本病毒性病害的发生情况对甘蔗常规杂交抗病育种中抗病亲本的选择具有重要指导意义。为此,本研究利用PCR及RT-PCR方法对中国175份甘蔗主要杂交亲本样品的甘蔗花叶病毒(sugarcane mosaic virus, SCMV)、高粱花叶病毒(sorghum mosaic virus, SrMV)、甘蔗线条花叶病毒(sugarcane streak mosaic virus, SCSMV)、甘蔗黄叶病毒(sugarcane yellow leaf virus, SCYLV)和甘蔗杆状病毒(sugarcane bacilliform virus, SCBV)等五种甘蔗主要病毒性病害进行了分子鉴定。结果显示,杂交亲本SCMV、Sr MV、SCSMV、SCYLV和SCBV的检出率分别为0、1.71%(3个)、0.57%(1个)、20.57%(36个)和69.71%(122个)。病毒复合侵染情况统计分析显示,175个样品中有42个未受这五种病毒中的任一种侵染,106个受单一病毒侵染,27个存在病毒复合侵染,复合侵染率为15.43%,其中复合侵染以SCYLV和SCBV侵染为主。本研究结果为中国甘蔗抗病杂交育种中抗病亲本的选择提供参考。 Knowing the current occurrence of viral diseases in major sugarcane hybrid parents in China is of great guiding significance for the selection of resistant parents in conventional cross-breeding of sugarcane. Therefore, PCR and RT-PCR methods were used to detect five major sugarcane viruses, including sugarcane mosaic virus (SCMV), sorghum mosaic virus (SrMV), sugarcane streak mosaic virus (SCSMV), sugarcane yellow leaf virus (SCYLV), and sugarcane bacilliform virus (SCBV) in 175 major sugarcane hybrid parents of China in this study. The results showed that the detection rates of SCMV, SrMV, SCSMV, SCYLV, and SCBV were 0, 1.71% (3 samples), 0.57% (1 sample), 20.57% (36 samples), and 69.71% (122 samples) respectively. Statistical analysis of virus mixed infection showed that 42 out of 175 samples were not infected by any of the five viruses, 106 were infected with a single virus, 27 were mixed infected with viruses, the mixed infection rate was 15.43%, of which SCYLV and SCBV were the main complication. These results provide a reference for the selection of resistant parents in disease-resistant cross-breeding of sugarcane in China.
