电针刺激对大鼠脊髓损伤后炎症反应的影响 被引量:19

Effects of Electroacupuncture on Inflammatory Response in Spinal Cord Injury of Rats

摘要 目的:观察不同配穴电针方式对脊髓损伤(SCI)大鼠IL-lβ,IL-6及IL-10表达的影响,探讨电针在脊髓损伤修复过程中的作用机制。方法:48只SD大鼠随机分为4组:假手术组(12例)、对照组(12例)、夹脊电针组(12例)、督脉电针组(12例)。术后第7天对各组大鼠进行BBB运动功能评分,并采用苏木精-伊红及Nissle染色观察组织细胞形态变化,采用实时荧光定量PCR法检测白介素-lβ(ILlβ),白介素-6(IL-6)和白介素-10(IL-10)mRNA的表达情况。结果:术后第7天对照组、夹脊电针组、督脉电针组BBB评分均低于假手术组(P<0.05),夹脊电针组、督脉电针组BBB评分均高于对照组(P<0.05);对照组、夹脊电针组、督脉电针组IL-lβ,IL-6及IL-10mRNA的表达含量均高于假手术组(P<0.05);夹脊电针组、督脉电针组IL-lβ,IL-6mRNA的表达含量均低于对照组(P<0.05);夹脊电针组、督脉电针组IL-10mRNA的表达含量均高于对照组(P<0.05);夹脊电针组与督脉电针组IL-lβ,IL-6及IL-10mRNA的表达差异无统计学意义(P>0.05)。结论:夹脊、督脉电针均可以改善大鼠SCI后下肢运动功能,降低IL-lβ和IL-6的表达,增加IL-10的表达,抑制SCI后炎症反应。 Objective:To observe the effect of electroacupuncture treatment(EA)at different acupoints on the expression of IL-1β,IL-6 and IL-10 in rats with spinal cord injury(SCI)and discuss the role and mechanism of electroacupuncture in the repair process of spinal cord injury.Methods:A total of 48 SD rats were randomly divided into 4 groups:sham operation group(n=12),control group(n=12),Jiaji EA group(n=12),and Du Meridian EA group(n=12).Scores of Basso Beattie Bresnahan(BBB)motor functions were assessed at 7 th days after operation.Histopathological changes were observed by HE and Nissle staining.The expressions of IL-1β,IL-6 and IL-10 mRNA were detected by quantitative real time-PCR(qRT-PCR).Results:The BBB scores of the control group,Jiaji EA group,and Du meridians EA group were lower than the sham operation group on the 7 th day after operation(P〈0.05).The BBB scores of Jiaji EA group and Du meridians EA group were higher than the control group(P〈0.05).The expression levels of IL-1β,IL-6 and IL-10 mRNA in the control group,Jiaji EA group and Du meridians EA group were higher than those in the sham operation group(P〈0.05).The expression levels of IL-1βand IL-6 mRNA in the Jiaji EA group and Du meridian EA group were lower than those in the control group(P〈0.05).The expression levels of IL-10 mRNA in Jiaji EA group and Du meridians EA group were high than those in the control group(P〈0.05).The expressions of IL-1β,IL-6 and IL-10 mRNA in Jiaji EA group and Du meridian EA group had no statistical significance(P〉0.05).Conclusion:Jiaji and Du Meridian EA could improve the motor function of the SD rats’ lower limb,reduce the expression of IL-1βand IL-6 and increase expression of IL-10 and inhibit the inflammatory response after SCI.
