摘要
以樱花品种白妙樱带芽茎段为材料,进行组织培养和快繁技术研究。结果表明:较适宜的启动培养基配方为:MS+6-BA(6-苄氨基嘌呤) 1. 0 mg·L^(-1)+NAA(萘乙酸) 0. 1 mg·L^(-1)。增殖阶段,先在MS+6-BA1. 0 mg·L^(-1)+NAA 0. 1 mg·L^(-1)+GA3(赤霉素) 5. 0 mg·L^(-1)中继代增殖15 d左右再转入MS+6-BA 1. 0mg·L^(-1)+2,4-D(2,4-二氯苯氧乙酸) 0. 2 mg·L^(-1)中,增殖效果较好,增殖系数高达4. 2。生根培养基较适宜的配方为:1/2MS+NAA 0. 5 mg·L^(-1),生根率为93. 63%。生根苗移栽至V珍珠岩∶V泥炭土∶V蛭石=1∶1∶1的基质中培养,室外炼苗30 d后,成活率达80%。
The stem of Cerasus serrulata var. lannesiana ‘Sirotae’ with buds were used as explants, sterilized and cultured on mediums with various combinations and concentrations of plant growth regulators for rapid progagation. The explants were cultured in a medium of MS+6-BA 1.0 mg·L -1 +NAA 0.1 mg·L -1 to induce shoots. The shoots were then subcultured in the medium of MS+6-BA 1.0 mg·L -1 +NAA 0.1 mg·L -1 +GA 3 5.0 mg·L -1 and then transferred onto the medium of MS+6-BA 1.0mg/L+2,4-D 0.2mg·L -1 for proliferation. The proliferation coefficient was 4.2 under these mediums. The plantlets were cultured on a rooting medium of 1/2MS+NAA 0.5 mg·L -1 , and the rooting rate was 93.63%. The rooted plantlets were transplanted in the substrate which was composed of pearlite, peat and vermiculite at a ratio of 1∶ 1∶ 1, and their survival rate was up to 80% after they hardened off outdoor for 30 d.
