亚砷酸钠对小鼠骨髓嗜多染红细胞微核率的影响

摘要 目的观察亚砷酸钠(Na As O_2)对小鼠骨髓嗜多染红细胞(PCE)微核形成的影响。方法昆明种小鼠雌、雄性小鼠各自随机分为6组,每组8只。以Na As O_2灌胃处理各组小鼠,以100 mg/kg环磷酰胺为阳性对照,以生理盐水为阴性对照,镜检计算嗜多染红细胞微核率,计算嗜多染红细胞/正染红细胞(NCE)比值。结果与阴性对照组比较,1.5625,3.125,6.25,12.5 mg/kg Na As O_2各剂量组均能增加雌、雄性小鼠骨髓PCE细胞的微核率,差异有统计学意义(P<0.05);且呈剂量依赖效应。各剂量Na As O_2处理组与阴性对照组的PCE/NCE比值比较,差异无统计学意义(P>0.05)。结论亚砷酸钠对小鼠骨髓嗜多染红细胞具有较强的促微核形成能力,且无性别差异。 Objective To evaluate the effect of sodium arsenite(NaAsO2)on the rate of micronucleus formation in bone marrow polychromatic erythrocytes(PCE)in mice.Methods First,48 male and female mice were randomly divided into 6groups,each group consisting 8 mice.Next,all groups were treated with NaAsO2for gastric lumen perfusion;Those treated with 100 mg/kg cyclophosphamide(CTX)were taken as the positive control group while those with normal saline were taken as the negative control.Microscopic observations of the micronucleus rate in PCE and PCE/normochromatic erythrocytes(NCE)ratio were then calculated.Results Compared with the negative control group,the groups of mice,both male and female,treated with Na As O2of 1.5625,3.125,6.25,and 12.5 mg/kg respectively,have shown increased the micronucleus rate in PCE cells.The difference was statistically significant(P〈0. 05).The increase of the micronucleus rate depended on the dosage.No significant difference in PCE/NCE ratio was found between the groups treated with Na As O2of various dosage and the negative control group(P〈0. 05).Conclusions Sodium arsenite can efficiently promote micronuclei formation in the bone marrow of mice with the subjects being either male or female.
