摘要
微量热技术是以测量细胞在代谢过程中能量变化规律为基础,通过细胞能量变化间接反应细胞活性,分析外界因素对其生长代谢影响的一种研究方法。微量热分析可以绘制生物体生长热谱图,通过建立数学模型,得到生长速率k,最大热功率Pm,传代时间tG及最小抑菌浓度(minimal inhibitory concentration, MIC)等热动力学参数。微量热技术是一项灵敏度高、测量准确的热分析技术,具有实时检测、在线分析、定量评估和动态描述等特点,已在生命科学领域得到了广泛的应用。本文主要讨论微量热技术在检测和比较原研与仿制抗生素药物中抑菌活性差异的应用可行性,为其在该领域的应用提供参考。
The microcalorimetry is a method used for recording of the heat produced by cells or bacterial populations over time. The change of the heat production indirectly reflects viability of cells or bacterial populations. Hence, microcalorimetric technique has increasingly been explored to study bacterial interactions with different factors in the medium culture. The process of bacterial growth can be monitored in real time by recording a graph of generated power over time. As a result, the power-time curves of bacteria can be continuously plotted in real time. It can provide abundant thermodynamic and kinetic parameters, such as the growth rate k, the generation time tG, the minimal inhibitory concentration (MIC), the maximum thermal power Pm, and so on. The aim of this study is to discuss the application of microcalorimetry to detect and compare anti-bacterial activity of original and generic antibiotics, providing information about the utilization ofmicrocalorimetry in this field.
