响应面法优化酶解海产小杂鱼及酶解液抗氧化性 被引量:4

To Optimize Enzymatic Hydrolysis of Small Marine Fish Protein and Antioxidant Activity of Its Hydrolysate by Response Surface Methodology

摘要 利用响应面法优化海产小杂鱼蛋白酶解工艺及酶解液抗氧化性研究。选用木瓜蛋白酶作为海产小杂鱼的酶解用酶,在单因素试验基础上,研究底物浓度、酶用量、酶解时间对酶解液的DPPH自由基清除率、肽回收率的影响。结果表明:以DPPH自由基清除率为响应值,得到的最佳酶解条件,为底物浓度4%,酶用量7%,酶解时间2.4 h,此时DPPH自由基清除率为60.82%;以肽回收率为响应值得到的最佳酶解条件,为底物浓度3%,酶用量7.20%,酶解时间2.6 h,此时肽回收率为55.31%。 Response surface methodology was used to optimize the enzymatic hydrolysis conditions of small marine fish protein and antioxidant activity of its hydrolysate was studied. Papain was used as the enzyme, and according to the results of single factor experiment, the effects of substrate concentration, enzyme amount and enzymolysis time on the scavenging rate of DPPH radical and peptide recovery were studied by response surface methodology. The results showed that the optimum enzymatic hydrolysis conditions were as follows: the substrate concentration 4%, the enzyme amount 7%, and the ertzymolysis time 2.4 h, and then the scavenging rate of DPPH radical was 60.82%; Under the substrate concentration 3%, enzyme dosage
