大鼠高脂饮食、颈动脉电击损伤复合因素所致动脉粥样硬化模型的研究 被引量:3

摘要 目的建立一个简单易行、价格低廉、有典型病理改变的大鼠动脉粥样硬化模型。方法通过对大鼠4、6、8、12周取材,进行颈动脉HE染色以及用ELISA方法测定大鼠血清TC、TG、HDL-C、LDL-C指标来判断模型制备情况。结果血脂:从实验4周末开始,假手术组、钳夹组、电击1组、电击2组、电击3组大鼠血清TC、TG和LDL-C均明显高于正常组(P <0.05),HDL-C较正常组明显降低(P <0.05)。病理变化:电击2组可以见到动脉内皮细胞脱落,大量平滑肌细胞移行至内膜下,血管壁向管腔内凸出,形成斑块,呈现典型的AS病理变化。结论利用高脂饮食、维生素D_2负荷加颈动脉电击损伤可以建立动脉粥样硬化模型。 Objective To establish a rat model of atherosclerosis that is simple, inexpensive and has typical pathological changes. Methods The rats were obtained at 4, 6, 8 and 12 weeks for carotid artery HE staining and ELISA for determination of TC, TG, HDL-C and LDL-C. Results Blood lipids: from the 4 th week of the experiment, serum TC, TG and LDL-C levels in sham group, clamp group, electroshock group 1, electroshock group 2 and electroporation 3 group were signifi cantly higher than those in normal group(P〈0.05). HDL-C was signifi cantly lower than the normal group(P〈0.05). Pathological changes: In the electroshock 2 group, arterial endothelial cells were observed to be shed, a large number of smooth muscle cells migrated to the subendocardial, and the blood vessel wall protruded into the lumen to form a plaque, showing typical AS pathological changes. Conclusion Atherosclerosis model can be established by using high-fat diet, vitamin D_2 loading and carotid electric shock injury.
