摘要
目的观察隔药饼灸对免疫抑制兔血细胞及免疫球蛋白水平影响的时效关系,阐明发挥效应的最佳时间。方法 90只新西兰大耳白兔采用随机数字表法分为9组,分别是空白组、模型即刻组、模型5天组、模型10天组、模型15天组、灸后即刻组、灸后5天组、灸后10天组和灸后15天组,每组10只。除空白组外,其余8组采用腹腔注射环磷酰胺建立免疫抑制兔模型。造模结束后第2天,灸法各组用六味地黄汤组方药饼,置于神阙、关元、足三里、脾俞、肾俞穴上施灸,隔日1次,共灸10次。空白组、模型即刻组、灸后即刻组实验结束后第2天,模型5天组和灸后5天组、模型10天组和灸后10天组、模型15天组和灸后15天组分别于正常饲养5、10、15天后检测血细胞系列[WBC、嗜中性粒细胞(NEU)百分数、淋巴细胞(LYM)百分数、单核细胞(MONO)百分数、嗜酸性粒细胞(EOS)百分数、嗜碱性粒细胞(BASO)百分数、红细胞(RBC)、血红蛋白(HGB)、红细胞压积(HCT)、血小板(PLT)],免疫球蛋白IgG、Ig M和补体C_3、C4水平。结果与空白组比较,模型即刻组、模型5天组、模型10天组和模型15天组WBC、RBC、HGB、HCT、PLT、NEU百分数,免疫球蛋白IgG、Ig M及补体C_3明显降低(P <0. 01),LYM百分数显著升高(P <0. 01),模型即刻组、模型5天组补体C4显著升高(P <0. 05)。与模型即刻组及模型5天组比较,灸后即刻组及灸后5天组WBC、RBC、HGB、HCT、PLT、NEU百分数,免疫球蛋白IgG、Ig M均明显升高(P <0. 05,P <0. 01),LYM百分数明显降低(P <0. 01);与模型即刻组比较,灸后即刻组补体C_3明显升高(P <0. 01);与模型5天组比较,灸后5天组补体C_3明显升高(P <0. 01),C4明显降低(P <0. 01)。与模型10天组比较,灸后10天组WBC、RBC、HGB、HCT、PLT,免疫球蛋白IgG、Ig M均明显升高(P <0. 05,P <0. 01),LYM百分数明显降低(P <0. 01)。与模型15天组比较,灸后15天组WBC、RBC、HGB、HCT及免疫球蛋白Ig M均明显升高(P <0. 01),LYM百分数明显降低(P <0. 01)。与灸后即刻组、灸后5天组比较,灸后10天组WBC、RBC、HGB、HCT、IgG、Ig M显著升高(P <0. 05,P <0. 01),LYM百分数明显降低(P <0. 01);与灸后10天组比较,灸后15天组WBC、RBC、HGB、HCT、PLT、免疫球蛋白IgG、Ig M均显著降低(P <0. 05,P <0. 01),LYM百分数显著升高(P <0. 05)。MONO、EOS、BASO百分数各组间比较,差异无统计学意义(P>0. 05)。结论隔药饼灸可以通过调节血细胞、免疫球蛋白和补体水平来调节环磷酰胺致免疫抑制兔的免疫功能,且调节作用存在一定的时效关系,在灸治结束后即刻起效,灸治10天效应最佳。
Objective To observe the time-effect of herb-cake-separated moxibustion on blood cell and immunoglobulin in immunosuppressive rabbits to explain the optimum time of exerting effect. Methods Totally 90 rab-bits were randomly divided into blank group,immediate-model group,5 days-model group,10 days-model group,15 days-model group,immediate after moxibustion group,5 days after moxibustion group,10 days after moxibustion group,15 days after moxibustion group with 10 rabbits in each group according to random digit table. Except the blank group,the other groups were given with intraperitoneal injection of cyclophosphamide to establish immunosuppressive rabbit model. Two days after modeling,moxibustion groups were treated with cake-separated moxibustion which herb-cake was made with Liuwei Dihuang Decoction Formula on Shenque( CV8),Guanyuan( CV4),Zusanli( ST36),Pishu( BL20) and Shenshu( BL23),one time every other day,a total of 10 times. Blank group,immediate-model group,immediate after moxibustion group,second days after the end of the experiment,5 days,10 days and 15 days of normal feeding in model 5 days-model group and 5 days after moxibustion group,10 days-model group and 10 days after moxibustion group,15 days-model group and 15 days after moxibustion group,the blood cells( including WBC,NEU%,LYM%,MONO%,EOS%,BASO5%,RBC,HGB,HCT,PLT),IgG,Ig M and C3,C4 level were detected in each time. Results Compared with blank group,the level of WBC,RBC,HGB,HCT,PLT,NEU%,IgG,Ig M and C3 in immediate-model group,5 days-model group,10 days-model group and 15 days-model group significantly decreased( P〈0. 01),while the LYM% decreased( P〈0. 01). Compared with blank group,the C4 of immediate-model group and 5 days-model group significantly increased( P〈0. 05). After the treatment,compared with immediate-model group and 5 days-model group,the level of WBC,RBC,HGB,HCT,PLT,NEU%,IgG,Ig M in immediate and 5 days after moxibustion groups increased( P〈0. 05,P〈0. 01) and the LYM%decreased( P〈0. 01). Compared with model-immediate group,the C3 of immediate after moxibustion group was increased( P〈0. 01); compared with 5 days-model group,the C3 of 5 days after moxibustion group was increased and C4 decreased( P〈0. 01). Compared with 10 days-model,the WBC,RBC,HGB,HCT,PLT,IgG and Ig M of 10 days after moxibustion group were increased( P〈0. 05,P〈0. 01) and the LYM% decreased( P〈0. 01). Compared with 15 days-model,the WBC,RBC,HGB,HCT and Ig M of 15 days after moxibustion group was increased( P〈0. 05,P〈0. 01) and the LYM% decreased( P〈0. 01). Compared with immediate and 5 days after moxibustion groups,WBC,RBC,HGB,HCT,IgG and Ig M of 10 days after moxibustion were significantly increased( P〈0. 05,P〈0. 01),the LYM% decreased significantly( P〈0. 05,P〈0. 01). Compared with the 10 days after moxibustion group,the WBC,RBC,HGB,HCT,PLT,IgG,Ig M of the 15 days after moxibustion group were significantly lower( P〈0. 05),and LYM% increased( P〈0. 05). The MONO%,EOS% and BASO% were not statistically significantly different in 9 groups( P〈0. 05). Conclusions Herb-cake-separated moxibustion could improve the immune function of rabbits under immunosuppressive state induced by cyclophosphamide by regulating the level of blood cells,immunoglobulin and complement,and it has a certain time-effect relationship. The effect is immediate after the moxibustion treatment,and the best effect accurs on the 10 th after the moxibustion treatment.
