摘要
目的观察昆明种雄性小鼠。肾脏中尿酸盐阴离子交换器1(urateanionexchanger1,URAT1)与miR-34a的相关性。方法40只SPF级昆明种雄性小鼠随机分成空白组和造模组,每组20只。造模组应用腺嘌呤联合氧嗪酸钾法制备高尿酸血症模型,测定小鼠血尿酸水平,应用荧光定量PCR法、Western印迹法检测肾脏URAT1基因表达水平及蛋白含量,应用荧光定量PCR法检测miR-34a基因表达量,进行Pearson相关分析。结果造模后造模组小鼠血尿酸、URAT1基因表达水平及蛋白含量较空白组升高,miR-34a基因表达量较空白组降低,差异有统计学意义(P〈0.05);血尿酸与URAT1为正相关关系(r=0.435),URAT1与miR-34a为负相关关系(r=-0.533)。结论miR-34a、URAT1、血尿酸之间可能存在相关性,上调miR-34。或可抑制URAT1基因表达,促进尿酸排泄,用于治疗高尿酸血症。
Objective To examine the relationship between urate anion exchanger 1 ( URAT1 ) and miR-34a in the kidney of Kunming male mice. Methods Forty Kunming male mice of specific pathogen-free (SPF) grade were randomly divided into control group and model group, with 20 mice in each group. Hyperuricemia model was established by use of adenine combined with potassium oxonate in the model group. Serum uric acid was measured in mice. Fluorescent quantitative PCR and Western blotting were used to detect the expression levels of URAT1 gene and protein in the kidney. Fluorescent quantitative PCR was used to measure the expression of miR-34a gene. Pearson cor- relation analysis was performed. Results The expression levels of uric acid, URAT1 gene and protein were much higher in the model group than those in the control group ( P 〈 0.05 ) . The expres- sion of miR-34a gene was lower in the model group than in the control group ( P 〈 O. 05 ) . Uric acid was positively correlated with URAT1 ( r = 0. 435 ) , and URAT1 was negatively correlated with miR-34a ( r = - 0. 533 ) . Conclusion There may be a correlation among miR-34a, URAT1, and uric acid. Up-regulation of miR-34a may inhibit the expression of URAT1 gene and promote the ex- cretion of uric acid. miR-34a may beused to treat hyperuricemia.
