摘要
目的:分析增液承气汤对肠道菌群失调大鼠神经肽分泌调节影响。方法:健康雄性SD大鼠40只,随机数字表法将大鼠分成四组,增液承气汤低剂量组、中剂量组、高剂量组及对照组,每组10只大鼠;制备肠道菌群失调大鼠模型,造模后低、中、高剂量组分别灌胃1ml剂量为0. 3ml/g、0. 2ml/g及0. 1ml/g增液承气汤,对照组每天灌胃1ml生理盐水,共灌胃7天;观察各组大鼠血清内EOS计数,肠道内细胞培养计数,免疫组化法检测各组大鼠肺及肠组织内血管活性肠肽(Vasoactive intestinal peptide,VIP)、降钙素基因相关肽(Calcitonin Gene Related Peptide,CGRP)和P物质(Substance P,SP)状况。结果:高、中、低剂量组大鼠双歧杆菌、乳酸杆菌低于对照组,高剂量组大鼠白色念珠菌、肠球菌及肠杆菌低于对照组,差异均有统计学意义(P<0. 05);低、中、高剂量组大鼠血清EOS计数低于对照组;低、中、高剂量组大鼠肺组织内CGRP、SP低于对照组,VIP高于对照组,差异有统计学意义(P<0. 05);低、中、高剂量组大鼠肠组织内CGRP、SP低于对照组,高剂量组大鼠VIP高于对照组,差异均有统计学意义(P<0. 05)。结论:增液承气汤可改善肠道菌群失调大鼠肠道菌群的失调状态,降低血清炎症因子含量,调节神经肽分泌状态。
Objective: The effect of Zengye Chengqi Decoction on the regulation of neuropeptide secretion in rats with dysbacteriosis was studied. Methods: 40 healthy male SD rats were selected from the Experimental Animal Center of our hospital. Rats were divided into four groups by random number table method. Zengqi Chengqi Decoction low-dose group,middle-dose group,high-dose group and controlled group,each group had 10 rats were prepared; A rat model of intestinal dysbacteriosis was prepared,and the doses of the low,middle,and high dose groups were each 0. 3 ml/g,0. 2 ml/g,and 0. 1 ml/g Zengqi Chengqi Decoction. The controlled group was given intragastric administration of 1 ml physiological saline every day for 7 days. The EOS counts in the serum of each group and the cell culture counts in the intestine were observed. The vasoactive intestinal peptides in the lung and intestine of each group were detected by immunohistochemistry. Vasoactive intestinal peptide( VIP),Calcitonin Gene Related Peptide( CGRP) and Substance P( SP) status. Results: The Bifidobacterium and Lactobacillus in the high-dose,medium-dose,and low-dose groups were lower than those in the controlled group. Candida albicans,Enterococcus and Enterobacter in the high-dose group were lower than those in the controlled group( P〈0. 05). Serum EOS counts in the low,medium and high dose groups were lower than those in the controlled group; CGRP and SP in the low,medium,and high dose groups were lower in the lung than in the controlled group,and VIP was higher than in the controlled group. The difference was statistically significant( P〈0. 05); The CGRP and SP in the intestine of the low-dose,medium-dose,and high-dose groups were lower than those in the controlled group. The VIP in the high-dose group was higher than that in the controlled group( P〈0. 05). Conclusion: Zengye Chengqi Decoction can improve the imbalance of intestinal microflora in rats with intestinal dysbacteriosis,reduce serum inflammatory cytokines,and regulate neuropeptide secretion.
