斑褐孔菌对室性心律失常大鼠保护作用的药效学研究 被引量:1

Pharmacodynamic study of Fuscoporia punctata on the protective function in ventricular arrhythmia rats

摘要 目的考察斑褐孔菌对乌头碱诱导大鼠室性心律失常的影响,为临床研究及应用提供可靠的试验依据。方法取健康SD大鼠32只,雌雄各半,随机分为4组,分别为斑褐孔菌低剂量组(0.27 g/kg)、中剂量组(0.81 g/kg)、高剂量组(1.35 g/kg)和对照组,每组各8只。斑褐孔菌各剂量组连续7 d灌胃给药,对照组灌同体积生理盐水。于第7 d给药后经颈静脉注射乌头碱以建立室性心律失常模型并监测心电图。结果与对照组相比,斑褐孔菌各剂量组大鼠心律失常出现时间均推迟,且低、中剂量组与对照组相比差异有统计学意义(P<0.05,P<0.01);其中中剂量组室性心律失常出现时间是对照组的6.2倍;与对照组相比,中、高剂量组出现心率失常的比例降低(P>0.05)。斑褐孔菌中、高剂量组均出现了窦性心律恢复大鼠,且中剂量组(37.5%)多于高剂量组(25.0%)。斑褐孔菌各剂量组死亡大鼠数均少于对照组;与对照组相比,斑褐孔菌中剂量组大鼠出现死亡的时间推迟。结论斑褐孔菌对乌头碱诱导的大鼠室性心律失常具有保护作用,可推迟室性心律失常发生时间及死亡时间,并有助于窦性心律恢复。 Objective To investigate the pharmacodynamics parameters of Fuscoporia punctata in the ventricular arrhythmia rats to provide a reliable experimental basis for prevention and treatment of arrhythmia. Methods Thirty-two healthy rats were divided into four groups, which were Fuscoporia punctata low-dose group(0.27 g/kg), medium-dose group(0.81 g/kg), high-dose group(1.35 g/kg) and control group. The rats of 3 Fuscoporia punctata groups were intragastrically administrated for 7 consecutive days(the control group was given the same volume of normal saline). Aconitine was injected via the jugular vein on the seventh day to establish a ventricular arrhythmia model and the electrocardiogram was monitored. Results Compared with control group, the occurrence time of arrhythmia in each Fuscoporia punctata group was delayed, and low-dose group and medium-dose group were significantly different from control group(P〈0.05, P〈0.01). The effect of medium-dose group was the most significant. Compared with control group, the proportions of arrhythmia in low-dose group and medium-dose group were decreased(P〈0.05). The rats in medium-dose and high-dose group had sinus rhythm recovery, and medium-dose group(37.5%) was higher than high-dose group(25.0%). The number of dead rats in each Fuscoporia punctata group was less than that of control group, and the time of death was delayed in the medium-dose group. Conclusion The Fuscoporia punctata has the protective effect on the ventricular arrhythmia in aconitine-induced rats, which can delay the occurrence and death time of ventricular arrhythmia, and restore the sinus rhythm.
