高糖环境对膀胱癌T24细胞Wnt5a表达的影响

Effect of high glucose environment on Wnt5a expression in bladder cancer T24 cells

目的探讨高糖环境对膀胱癌T24细胞Wnt5a表达的影响。方法将膀胱癌T24细胞分为空白组、对照组、高糖1组、高糖2组及高糖3组,分别在含5. 5 mmol/L葡萄糖、5. 5 mmol/L葡萄糖+25 mmol/L甘露醇、10 mmol/L葡萄糖、20 mmol/L葡萄糖、30 mmol/L葡萄糖的杜氏改良伊格尔培养基(DMEM)完全培养基内培养,48 h后检测各组T24细胞Wnt5a mRNA及蛋白的相对表达水平。结果空白组与对照组Wnt5a mRNA及Wnt5a蛋白相对表达量比较,差异均无统计学意义(P> 0. 05),高糖1组、高糖2组、高糖3组Wnt5a mRNA及Wnt5a蛋白相对表达量均高于空白组与对照组,且依次升高(均P <0. 05)。结论体外高糖环境可提高膀胱癌T24细胞Wnt5a的表达,从而促进膀胱癌的发生发展。

Objective To investigate the effect of high glucose environment on the expression of Wnt5a in bladder cancer T24 cells. Methods The bladder cancer T24 cells were cultured in the complete medium of Dulbecco′s modified Eagle medium（DMEM） with different concentrations of glucose,5.5 mmol/L glucose（blank group）,5.5 mmol/L glucose＋25 mmol/L mannitol（control group）, 10 mmol/L （high glucose 1 group）,20 mmol/L（high glucose 2 group） and 30 mmol/L（high glucose 3 group）.And 48 hours later,the relative expression levels of Wnt5a mRNA and protein in T24 cells were detected in each group. Results There was no statistically significant difference in the relative expression of Wnt5a mRNA or Wnt5a protein between the blank group and the control group（ P 〉0.05）.The relative expressions of Wnt5a mRNA and Wnt5a protein in the high glucose 1 group,high glucose 2 group and high glucose 3 group,which increased in turn,were higher than those in the blank group and control group（all P 〈0.05）. Conclusion The high glucose environment in vitro can increase the expression of Wnt5a in bladder cancer T24 cells,thus promoting the occurrence and progression of bladder cancer.
