Determination of chlorhexidine acetate in herbal antibacterial agents

摘要 目的建立快速检测植物草本抑菌剂中醋酸氯己定含量的高效液相色谱方法。方法样品经硫酸溶液消除拮抗作用,纯水稀释后超声提取,提取液过滤后经反相液相色谱分离,二极管阵列检测器检测。结果醋酸氯己定浓度在10μg/ml~150μg/ml时,线性相关系数r> 0. 999 9。对样品进行8次平行测定,相对标准偏差为1. 35%;在低、中、高(分别对应50%、100%、150%样品含量) 3个水平加标浓度下,回收率在94. 66%~98. 41%。结论该方法简单、快速、准确,适用于植物草本抑菌剂中的醋酸氯己定含量测定。 Objective To develop a method for the fast determination of chlorhexidine acetate in herbal antibacterial agents by high performance liquid chromatography with ultraviolet detector( HPLC - UV). Methods The sample was mixed with sulfuric acid solution to eliminate antagonism by ultrasonic extraction. After dilution with pure water, the mixture was extracted by ultrasonication, filtered, separated by reversed - phase liquid chromatography, and detected by a diode array detector. Results It showed that chlorhexidine acetate had a good linearity ( r 〉 0. 999 9) in the range of 10 μg/ml - 150 μg/ml. The relative standard deviation of ehlorhexidine acetate in sample was 1.35% for eight parallel determinations. The spiking recovery ranged from 94.66% to 98.41% at three different spiked levels ( 50% , 100% , 150% ). Conclusion The established method is simple, rapid and has good selectivity and stability. It is suitable for the determination of chlorhexidine acetate in different herbal antibacterial agents.
