胃癌患者外周血中性粒细胞凋亡、活化状态及功能研究

Neutrophil apoptosis,activation condition and the function of peripheral blood in gastric cancer patients

目的观察胃癌患者外周血中性粒细胞凋亡、活化状态; ROS产生能力;对胃癌细胞MGC-803的杀伤能力。方法 20例来自作者医院的胃癌初发患者,以及20例健康献血者。在提取中性粒细胞之后,用流式细胞技术检测2组中性粒细胞的Caspase3和CD66b的表达情况;用ELISA法检测2组中性粒细胞的活性氧含量;与胃癌细胞株MGC-803共培养检测2组中性粒细胞的肿瘤细胞杀伤能力。结果 Caspase3在胃癌患者外周血中性粒细胞中的表达(13. 05%±14. 46%)较健康人群(48. 08%±35. 31%)明显降低,差异有统计学意义(P <0. 01);但CD66b的表达在两者中差异无统计学意义(P> 0. 05)。胃癌患者外周血中性粒细胞活性氧含量明显低于健康对照组(P <0. 001),且对肿瘤的杀伤能力降低。结论胃癌患者外周血中性粒细胞的凋亡率降低,但是活化程度未有变化。胃癌患者外周血中性粒细胞活性氧释放能力降低,且杀伤能力降低。

Objective To observe the apoptosis and activation state of neutrophils in peripheral blood of gastric cancer patients, the ability of ROS production, and the killing ability of MGC - 803 in gastric cancer cells. Methods 20 patients with primary gastric cancer and 20 healthy blood donors came from Lishui People＇s Hospital. After extracting neutrophils, flow cytometry was used to detect the expression of Caspase3 and CD66b in two groups of neutrophils. Then active oxygen content in two groups of neutrophils was detected by ELISA. The gastric cancer cell line MGC - 803 was co - cultured to detect the killing ability of neutrophils between the two groups. Results The expression of Caspase3 in peripheral blood neutrophils of patients with gastric cancer （13.05% _＋ 14.46% ） was significantly lower than that of the healthy population （48.08% ＋ 35.31% ）, with the difference statistically significant （P 〈0.01 ）; but there was no statistical significance on the difference of CD66b expression. The active oxygen content of neutrophils in peripheral blood of gastric cancer patients was significantly lower than that of the healthy control group （ P 〈 0. 001 ）, and the killing ability of the tumor was reduced. Conclusion The apoptosis rate of neutrophils in peripheral blood of gastric cancer patients decreased, but the activation degree didn＇t change. The release of reactive oxygen species from neutrophils in peripheral blood of gastric cancer patients was reduced, and the ability to kill the neutrophils was reduced.
