摘要
目的 探讨HLA-DQB1基因多态性与河南汉族女性宫颈癌易感性及人乳头瘤病毒(HPV)感染的关系.方法 分别采集于2016年5月至2017年9月在郑州人民医院就诊的117例宫颈癌患者和110例子宫良性病变患者的静脉血和宫颈组织,分别提取其全基因组DNA和HPV核酸,分别采用聚合酶链式反应序列特异性引物(PCR-SSP)和HPV分子导流杂交技术进行HLA-DQB1基因分型和HPV分型,并进行统计学分析.结果 117例宫颈癌患者中,HPV感染率为79.5%,且HPV16是主要致病亚型;110例子宫良性病变患者中,HPV感染率为9.1%.宫颈癌患者中HLA-DQB1﹡03等位基因出现的频率明显高于子宫良性病变患者,HLA-DQB1﹡06等位基因出现的频率低于子宫良性病变患者,差异有统计学意义(P〈0.05);HLA-DQB1﹡03等位基因在HPV16阳性组中的频率(40.20%)明显高于HPV16阴性组(20.45%,P〈0.05).结论 HLA-DQB1﹡03等位基因可能是河南汉族女性宫颈癌发生的易感基因,而HLA-DQB1﹡06等位基因可能是其保护基因;携带HLA-DQB1﹡03等位基因的女性更容易感染HPV16病毒,增加宫颈癌的发生风险.
Objective To investigate the relationship between HLA-DQB1 gene polymorphism and susceptibility to cervical cancer and human papiullama virus ( HPV ) infection in Han women in Henan Province .Methods The venous blood and the cervical tissue were collecting from 117 patients with cervical cancer and 110 patients with benign uterine lesions , all of which were treated in Zhengzhou People's Hospital from May 2016 to September 2017;the genomic DNA and the HPV nucleic acid were separately extracted . The HLA-DQB1 typing and the HPV typing were respectively performed by PCR-SSP and HPV molecular flow-diversion hybridization , and then the results were analyzed statistically .Results Among the 117 patients with cervical cancer , the rate of HPV infection was 79.5%, and HPV16 was the main pathogenetic subtype;while the rate of HPV infection in patients with benign uterine lesions was 9.1%.The occurrence frequency of HLA-DQB1﹡03 allele in cervical cancer patients was significantly higher than that in patients with benign uterine lesions , but the occurrence frequency of HLA-DQB1﹡06 allele was lower than that in patients with benign uterine lesions ( P〈0.05 ) .The occurrence frequency of HLA-DQB1﹡03 allele in HPV16-positive group ( 40.20%) was significantly higher than that in HPV 16-negative group ( 20.45%, P〈0.05 ) .Conclusions The HLA-DQB1﹡03 allele may be a susceptible gene of cervical cancer in Han women in Henan Province , while the HLA-DQB1﹡06 allele may be the protective gene .Women with HLA-DQB1﹡03 allele were more likely to be infected with HPV16 virus and have high risk of cervical cancer .
